摘　　要：【目的】探究蚯蚓提取物对葡聚糖硫酸钠(dextran sulfate sodium,DSS)诱导的小鼠溃疡性结肠炎(ulcerative colitis,UC)的影响,为蚯蚓用于UC潜在治疗药物提供试验依据。【方法】C57BL/6小鼠自由饮用3.5%DSS溶液7 d并灌胃不同浓度的蚯蚓提取物,每日监测小鼠精神状态、体质量和大便性状;于第10天解剖,取脾脏和结肠,制作结肠组织石蜡切片并进行苏木精—伊红染色;以荧光定量PCR检测结肠组织中肿瘤坏死因子α (TNF-α)、白细胞介素6 (IL-6)、白细胞介素1β (IL-1β)和白细胞介素10 (IL-10)的m RNA表达量;以ELISA检测结肠组织中闭锁小带蛋白1 (ZO-1)、黏蛋白2 (Muc2)和咬合蛋白(occludin)的表达量。【结果】蚯蚓提取物组能减轻小鼠体质量下降程度,降低小鼠疾病活动指数评分,减少炎性细胞浸润程度,下调IL-6、IL-1β和TNF-α的mRNA表达,上调IL-10的mRNA表达以及ZO-1、occludin和Muc2的蛋白表达量。【结论】蚯蚓提取物对DSS诱导的小鼠UC具有缓解作用,小鼠结肠组织炎性损伤和UC症状减轻。蚯蚓提取物可保护结肠黏膜的完整性和改善肠黏膜屏障功能。[Purpose]To investigate the effects of earthworm extracts on the dextran sulfate sodi-um(DSS)-induced ulcerative colitis(UC)in mice,providing experimental evidence for earthworm extracts as potential therapeutic drugs for UC.[Methods]C57BL/6 mice drank 3.5%DSS solution freely for seven days and gavaged with different concentrations of earthworm extracts.The psychosis body weight,and stool characteristics were monitored daily.Dissection was performed on the 10th day,and the spleen and colon were sampled.Colon pathological sections were made by hematoxylin-eosin staining,and the mRNA expression level of tumor necrosis factorα(TNF-α),interleukin 6(IL-6),interleukin 1β(IL-1β)and interleukin 10(IL-10)were detected by qRT-PCR;zonula occludens protein 1(ZO-1),mucin 2(Muc2),and occludin protein expression in colon tissue was detected by ELISA.[Results]The earthworm extracts reduced body weight loss,disease activity index score and the degree of inflammatory cell infiltration in mice,down-regulated the mRNA expression level of IL-6,IL-1βand TNF-α,and up-regulated the mRNA expression of IL-10 and the protein expres-sion of ZO-1,occludin and Muc2.[Conclusion]Earthworm extract has a relieving effect on DSS-induced UC in mice,relieved the inflammatory damage of colon tissue and the symptoms of UC,pro-tecting the integrity of the colonic mucosa and improving the intestinal mucosal barrier function.
