摘  要:【目的】研究镉(Cd)胁迫下不同钙(Ca^(2+))浓度处理对深色有隔内生真菌——嗜鱼外瓶霉生长、矿质营养和Cd含量的影响。【方法】设置5个Ca^(2+)浓度处理(0.045、0.225、0.450、2.250和4.500 mmol/L),测定100 mg/L Cd胁迫下,嗜鱼外瓶霉的菌丝干质量、孢子数、培养液pH以及菌丝Cd、Ca、氮(N)、镁(Mg)、硫(S)和磷(P)的含量,分析菌丝干质量、菌丝Cd含量、培养液Ca含量与菌丝矿质营养含量的相关性。【结果】Cd胁迫下,0.225 mmol/L Ca^(2+)处理的嗜鱼外瓶霉菌丝干质量和孢子数最大;0.450 mmol/L Ca^(2+)处理下菌丝中Cd含量最高。不同浓度Ca^(2+)可显著影响嗜鱼外瓶霉菌丝Mg、N、P、Ca和S元素的含量;相关性分析表明:不同矿质元素对菌丝干物质累积和Cd、Ca元素的累积作用影响有较大差异;Cd胁迫下菌丝Cd含量与Mg和P的吸收有协同作用;培养液外源添加Ca可改善菌丝对营养液中Ca和S元素的吸收。【结论】嗜鱼外瓶霉菌丝培养过程中,添加适量Ca^(2+)可促进菌丝生长,且以0.225 mmol/L Ca^(2+)最有利于菌丝和孢子生长,也比较利于菌丝累积N、P、Mg和S。为了减轻菌丝对Cd的吸附,可适量添加P和Mg;不适宜的矿质元素浓度反而会毒害菌丝生长。[Purpose]To study the effects of different calcium(Ca^(2+))concentrations on the growth,mineral nutrition and cadmium(Cd)content of a dark septate endophytic fungus,Exophiala pis-ciphila,under Cd stress.[Methods]Five Ca^(2+)concentration levels(0.045,0.225,0.450,2.250 and 4.500 mmol/L)were set as treatments in the experiment to determine the mycelial dry weight,spore number,pH value of culture solution and the contents of Cd,Ca,nitrogen(N),magnesium(Mg),sul-fur(S)and phosphorus(P)of E.pisciphila under 100 mg/L Cd stress.And the correlations among the above-mentioned indexes were analyzed.[Results]Under Cd stress,0.225 mmol/L Ca^(2+)treatment resulted in the highest mycelial dry weight and spore number of E.pisciphila;the Cd content in the mycelium was the highest under 0.450 mmol/L Ca^(2+)treatment.Different concentrations of Ca^(2+)could significantly affect the content of Mg,N,P,Ca,and S elements in the mycelium of E.pisciphila;cor-relation analysis showed that there were significant differences in the effects of different mineral ele-ments on the accumulation of dry weight,Cd and Ca in mycelium;there was a synergistic effect between Cd content in mycelium and the absorption of Mg and P under Cd stress;adding exogenous Ca to the culture medium could improve the mycelium’s absorption of Ca and S elements in the nutri-ent solution.[Conclusion]During the cultivation process of E.pisciphila,adding an appropriate amount of Ca^(2+)can promote mycelial growth.The addition of 0.225 mmol/L Ca^(2+)is most conducive to the growth of mycelium and steamed buns,and also conducive to the accumulation of N,P,Mg,and S.In order to reduce the adsorption of Cd by mycelium,an appropriate amount of P and Mg can be added.Inappropriate concentrations of mineral elements can actually poison mycelium growth.
