摘  要:目的:通过比较分析小鼠骨髓来源的间充质干细胞(mBMSCs)和人脐带来源的间充质干细胞(hUCMSCs)的生物学共性特征,为选择hUCMSCs作为移植细胞,开展临床前体内药效评价提供理论依据。方法:采用“全骨髓贴壁法”联合“红细胞裂解法”,分离、纯化得到mBMSCs,再与已有的hUCMSCs进行比较,采用荧光倒置显微镜下观察两者的形态学特点,采用cck8法比较两者增殖活性,采用流式细胞术分析两者表面标志物及干性标志物表达。结果:mBMSCs和hUCMSCs的形态特征高度相似,均呈螺旋状贴壁生长,形态均一,多成梭形。但mBMSCs传代至7代以上,则形态开始改变,有分化趋势,而hUCMSCs传至10代以上,仍稳定生长。mBMSCs和hUCMSCs的表面标志物表达高度相似,CD90及CD105表达率均≥95%,CD45及CD34表达率均≤2%。此外,与mBMSCs相比,hUCMSCs展示了更高的体外增殖活性,并且干性标志物Oct-4、Sox2和Nanog的表达量更高,这可能与细胞来源及体外培养条件有关。结论:mBMSCs和hUCMSCs的形态学特征及表面标志物表达有高度相似性,但hUCMSCs体外增殖活性和干性标志物表达量相对较高。该研究为以hUCMSCs作为移植细胞开展临床前体内药效评价提供了部分科学依据。Aims:This paper aims to provide the scientific basis for the application of human umbilical cord mesenchymal stem cells(hUCMSCs)as transplanted cells in preclinical in vivo efficacy evaluation.The common features of mouse bone marrow mesenchymal stem cells(mBMSCs)and hUCMSCs were compared by an analysis of the biological characteristics.Methods:mBMSCs were isolated and purified by the"whole bone marrow adherent method"combined with the"red cell lysis method"and then compared with the existing serum-free human umbilical cord mesenchymal stem cells(hUCMSCs).The morphological characteristics of mBMSCs and hUCMSCs were observed and compared under the inverted fluorescence microscope.The proliferative activity of the two MSCs was compared by using the cck8 method;and the expressions of the surface markers and stemness markers of the two MSCs were analyzed by flow cytometry.Results:The results showed that the morphological characteristics of mBMSCs and hUCMSCs were highly similar.The two MSCs grew in a spiral shape and were attached to the wall,with a uniform shape,mostly forming a fusiform shape.However,when the mBMSCs were passaged to more than the 7th generations,their morphology began to change and showed a tendency to differentiate.But hUCMSCs could still grow steadily after they were passaged to more than 10 generations.The expressions of the surface markers CD90,CD105,CD45 and CD34 were also very similar between mBMSCs and hUCMSCs.The expression rates of CD90 and CD105 of the two MSCs were greater than or equal to 95%;and those of CD45 and CD34 were less than or equal to 2%.In addition,compared with mBMSCs,hUCMSCs showed higher proliferative activity and higher expression levels of the stemness markers Oct-4,Sox2 and Nanog.It could be related to different cell resources and the in vitro culture condition.Conclusions:These results indicated that hUCMSCs was similar to mBMSCs in morphological characteristics and surface marker expresions.But hUCMSCs showed higher proliferative activity and stemness.This study prov
