Investigation on Orchid Viral Diseases in Shanghai Chenshan Botanical Garden

摘　　要：采用DAS-ELISA、免疫层析胶体金试纸条、RT-PCR和小RNA深度测序检测方法,对上海辰山植物园53属124份兰花样品的病毒种类、被侵染兰花种类、症状进行了全面调查。结果表明:所测样品中仅含建兰花叶病毒(CymMV)和齿兰环斑病毒(ORSV),CymMV、ORSV和复合侵染的感染率分别46%、31%和23%,症状主要为黄绿斑驳嵌纹、黑色凹陷坏疽、叶肉皱缩变色、白拉丝等,感染CymMV、ORSV病毒的兰花分别有29、19个属,其中石斛、卡特兰、树兰、文心兰等属兰花病毒病的感染率较高,并据此提出了相应的防控建议。The virus species,hosts and symptoms of 53 genera 124 orchid samples in Shanghai Chenshan Botanical Garden were investigated by DAS-ELISA,complex gold immunochromatography assay,RT-PCR and microRNA deep sequencing.The results showed that the tested samples only contain Cymbidium mosaic virus(CymMV) and Ornithogalum ringspot virus(ORSV).The infection rates of CymMV,ORSV and compound infection were 46%,31% and 23%.The main symptoms are yellow-green mottled,black depression or convex gangrene,mesophyll shrinkage,distortion,discoloration and necrosis,etc.There were 29 and 19 genera of orchids infected with CymMV and ORSV,respectively.Among them,the infection rate of orchid virus diseases of Dendrobium,Cattleya,Epidendrum,and Oncidium were higher.Accordingly,corresponding prevention and control suggestions are put forward.
