摘  要:嗜肺军团菌是一种寄生在真核细胞内的革兰氏阴性致病菌,其致病性主要是依靠IVB型分泌系统分泌的330多种效应蛋白,效应蛋白之间彼此协作共同干扰真核宿主细胞的各种细胞活动.嗜肺军团菌效应蛋白Lpg1972与真核细胞Ccr4-Not复合物中的CNOT7亚基存在相互作用,而Ccr4-Not复合物是真核细胞中调控mRNA降解的关键蛋白。基于此通过分子生物学的手段验证了它们之间的相互作用,设计共表达载体纯化了复合物蛋白,并利用AlphaFold2对复合物的结构和相互作用进行了预测分析,为后续研究奠定了基础。Legionella pneumophila is a Gram-negative pathogenic bacterium that parasitizes within eukaryotic cells.Its pathogenicity primarily relies on the secretion of over 330 effector proteins through the Type IVB secretion system.These effector proteins collaborate to collectively interfere with various cellular activities of the eukaryotic host cell.The Legionella pneumophila effector protein Lpg1972 has been identified to interact with the CNOT7 subunit of the Ccr4-Not complex in eukaryotic cells.The Ccr4-Not complex predominantly regulates mRNA degradation in eukaryotic cells.To validate this interaction,molecular biology techniques were employed,involving the design of a co-expression vector to purify the protein complex.The complex protein was successfully obtained and its structure and interaction were predicted using AlphaFold2.These analyses lay a foundational basis for subsequent research,establishing a comprehensive understanding of the structural and functional aspects of the identified interaction.
