养精胶囊通过调节自噬提高睾丸间质细胞睾酮合成功能的研究

摘  要:目的 探讨养精胶囊促进睾丸间质细胞合成睾酮(T)的具体机制。方法 将雄性BALB/c小鼠饲养至18月龄,以生理盐水和不同剂量养精胶囊混悬液灌胃处理30天。ELISA法检测小鼠血清T水平;Western blot检测小鼠睾丸组织Beclin-1和LC3蛋白的表达水平;透射电镜观察小鼠睾丸间质细胞的自噬情况。结果 养精胶囊灌胃处理后,老年小鼠的血清T水平增加,且呈剂量依赖性,养精胶囊低、中、高剂量组小鼠的血清T与老年对照组相比,均有显著差异(P<0.01);养精胶囊低、中、高剂量组小鼠睾丸组织中的Beclin-1和LC3 II/I均显著高于老年对照组(P<0.05);透射电镜结果显示,与老年对照组相比,养精胶囊低、中、高剂量组小鼠睾丸组织中间质细胞内自噬小体明显增多,尤其是养精胶囊高剂量组。结论 养精胶囊可以通过调控睾丸间质细胞的自噬,从而提高其合成T的功能。Objective To explore the mechanism of Yangjing capsule promoting the testosterone synthesis in Leydig Cells.Methods Male BALB/c mice were raised to 18 months old and treated with saline and different doses of Yangjing capsule by gavage for 30 days.The serum level of testosterone was measured by ELISA,the protein expression levels of Beclin⁃1 and LC3 in testicular tissue of mice were assayed by Western blot,autophagy in Leydig cells of mice was observed by transmission electron microscopy.Results After intragastric administration of Yangjing capsule,the serum testosterone level of old mice increased in a dose⁃dependent manner,compared with the elderly control group,there were significant differences in serum testosterone levels in mice among the low,medium and high dose of Yangjing capsule groups(P<0.01).The Beclin⁃1 and LC3 II/I levels in the testicular tissue of mice in the low,medium and high dose of Yangjing capsule groups were significantly higher than those in the control group(P<0.05).The transmission electron microscopy results showed that compared with the control group,the number of autophagosomes in Leydig cells of testicular tissue was significantly increased of the low,medium and high dose of Yangjing capsule groups,especially in the high⁃dose of Yangjing capsule group.Conclutions Yangjing capsule can improve the function of testosterone synthesis by regulating autophagy in Leydig cells.
