摘 要:目的:探究石斛多糖(Dendrobium sonia polysaccharide,DSP)对关节炎(collagen induced arthritis,CIA)模型小鼠的保护作用及其与氧化应激的关系。方法:将C57BL/6小鼠随机分为对照组、CIA组、L-DSP组、M-DSP组、H-DSP组。使用低、中、高浓度的DSP处理CIA模型小鼠。HE染色观察各组小鼠后肢组织病理改变,ELISA检测血清TNF-α、PGE_(2)、4-HNE水平,生化检测踝关节组织GSH、GSSG和NADP^(+)/NADPH水平,Western blot检测右肢踝关节组织IDH2蛋白表达水平。结果:相比对照组,CIA组小鼠后肢肿胀明显,滑膜增生、淋巴细胞浸润明显,TNF-α、PGE_(2)、4-HNE、GSSG、NADP^(+)/NADPH水平升高(P<0.05),GSH和IDH2 mRNA和蛋白水平降低(P<0.05);相比CIA组,DSP组小鼠后肢肿胀减轻,滑膜增生、淋巴细胞浸润减少,TNF-α、PGE_(2)、4-HNE、GSSG、NADP^(+)/NADPH水平降低(P<0.05),GSH和IDH2 mRNA和蛋白水平升高(P<0.05),且均呈DSP剂量依赖性。结论:DSP可通过减少氧化应激反应减轻CIA小鼠的关节肿胀程度和炎症反应。OBJECTIVE To explore the protective effect of Dendrobium sonia polysaccharide(DSP) in collagen induced arthritis(CIA) model mice and the relationship with oxidative stress.METHODS Mice were randomly divided into control group,CIA group,and L-DSP,M-DSP and H-DSP groups.CIA model mice were treated with low,medium and high concentrations of DSP.HE staining was used to observe the pathological changes of hind limbs of mice in each group.Serum levels of TNF-α,PGE_(2) and 4-HNE were determined by ELISA assay.The levels of GSH,GSSG and NADP~+/NADPH in ankle joint were detected by biochemical kits.The protein expression level of IDH2 was detected by Western blot.RESULTS Compared with the control group,in CIA group,the swelling of hind limbs,synovial hyperplasia and lymphocyte infiltration were obvious in mice,the levels of TNF-α,PGE_2,4-HNE,GSSG and NADP~+/NADPH were increased,and the levels of GSH and IDH2 mRNA and protein were decreased.Compared with CIA group,in DSP groups,the swelling of hind limbs,synovial hyperplasia and lymphocyte infiltration in mice were reduced,the levels of TNF-α,PGE_2,4-HNE,GSSG and NADP~+/NADPH were decreased,and the levels of GSH and IDH2 mRNA and protein were increased,and all of the effects were in DSP dose-dependent manner.CONCLUSION DSP can reduce the degree of joint swelling and inflammatory reaction in CIA mice by reducing oxidative stress.
