摘 要:目的探讨高癌家系气虚质鼻咽癌初诊患癌组织中α-enolase的表达活性水平及其临床病理意义。方法分别收集高癌家系中气虚质鼻咽癌初诊患者、鼻咽黏膜慢性炎症患者各9例,非高癌家系气虚质鼻咽癌初诊患者12例的鼻咽黏膜组织,Real Time PCR分别检测ENO1 mRNA表达活性,Western blotting检测ENO1蛋白表达水平,比较分析其组间差异及其临床病理意义。结果高癌家系组、非高癌家系气虚质鼻咽癌患者组、健康人鼻咽组织中ENO1 mRNA的△Ct分别为2.45±0.42,3.47±0.28,4.49±0.51;2^(-△△Ct)值分别为4.09±1.27,1.97±0.38,1.00±0.30;各组ENO1蛋白相对表达量依次为2.94±0.81、1.73±0.53、1.27±0.25;ENO1 mRNA及其蛋白表达水平组间差异均具有统计学意义(P<0.05)。结论高癌家系气虚质初诊鼻咽癌患者ENO1 mRNA及蛋白表达水平的明显上调具有显著的临床病理意义,对高癌家系气虚质鼻咽癌患者早期筛查及诊断可能具有较好的临床应用价值。Objective To observe whetherα-enolase(ENO1)is differential expression protein in patients with nasopharyngeal carcinoma who are Qi-deficiency constitution in high cancer families.Methods Collect nasopharyngeal mucosal tissues from 9 newly diagnosed patients with Qi deficiency nasopharyngeal carcinoma in high cancer families,9 healthy individuals,12 case with Qi deficiency of nasopharyngeal carcinoma from scatteredly distributed patients,at the same time collecting 9 cases nasopharyngeal tissue from chronic inflammation of nasopharyngeal mucosa patients,to extract total RNA and protein respectively.Results The results of ENO1 mRNA using Real Time PCR showed that the△Ct of ENO1 mRNA in nasopharyngeal tissue from Nasopharynx tissue of a healthy person,Qi deficiency of nasopharyngeal carcinoma from scatteredly distributed patients and from high cancer families were 4.49±0.51,3.47±0.28,2.45±0.42,and the 2^(-△△Ct)were 1.00±0.30,1.97±0.38,4.09±1.27 respectively;the relative expression of ENO1 mRNA in Qi deficiency of nasopharyngeal carcinoma from scatteredly distributed patients and from high cancer families were 1.97 times and 4.09 times than that of nasopharyngeal tissue from chronic inflammation of nasopharyngeal mucosa patients.The differences among three groups were all significant(P<0.05).Conclusion The expression of ENO1 protein was significant up-regulated in patient with Qi deficiency of nasopharyngeal carcinoma from high cancer families and from scatteredly distributed patients.ENO1 is expected to be early diagnosis and screening markers in patient with Qi deficiency constitution of nasopharyngeal carcinoma of high cancer families.
