摘 要:目的:探究小鼠颈动脉斑块程度与血管壁剪切应力(wall shear stress,WSS)的关系,以及对心脏功能的影响。方法:将40只,周龄相仿且敲除载脂蛋白E(ApoE-/-)的雄性小鼠(遗传背景为C57BL/6J)分为四组,分别喂养普通饮食、高脂饮食、注入血管紧张素II(Ang II)的高脂饮食、注入Ang II和Periostin重组腺病毒的高脂饮食,4周后对小鼠进行超声检查和病理切片苏木精-伊红染色。结果:与正常组小鼠相比,高脂饲料喂养的A、B、C组小鼠TC、TG、LDL逐渐升高,而HDL逐渐减低,差异有统计学意义(P<0.05);与正常组小鼠相比,高脂饲料喂养的A、B、C组小鼠的WSS值降低,A组降低了1.5%,B组降低了23.2%,C组降低了35.4%,随着斑块程度加重WSS随之降低;与正常组小鼠相比,高脂饲料喂养的A、B、C组小鼠的LVESD、LVEDD、左心室收缩末期容积、左心室舒张末期容积、每搏量、心输出量升高,而LVEF、短轴缩短率降低比较,差异有统计学意义(P<0.05)。结论:斑块程度与WSS间为较强的负相关性,低剪切应力诱导ApoE-/-小鼠颈动脉粥样硬化的形成,并且Periostin正向调控Ang II抑制脂质代谢的作用。Objective:To investigate the relationship of the degree of carotid plaque with wall shear stress(WSS)and the effect on the heart in mice.Methods:Male ApoE gene knockout mice on C57BL/6J background(n=40)were divided into four groups,which were fed normal diet,high-fat diet,high-fat diet infused with angiotensin Ⅱ(Ang Ⅱ),high-fat diet infused with Ang Ⅱ and Periostin recombinant adenovirus.After 4 weeks,mice were examined by ultrasound and pathological sections were stained with hematoxylin and eosin.Results:Compared with mice in normal group,TC,TG and LDL of mice in groups A,B and C fed with high fat diet gradually increased,while HDL gradually decreased(P<0.05).Compared with the normal group,the WSS values of the mice in the A,B,and C groups fed with high-fat diet were decreased.The WSS values of high-fat diet group A,B and C were decreased by 1.5%,23.2%and 35.4%,respectively.WSS decreased with the severity of plaque.Compared with the normal group,the LVESD,LVEDD,left ventricular end-systolic volume,left ventricular end-diastolic volume,stroke volume and cardiac output in the high-fat diet group A,B and C were increased,while the left ventricular ejection fraction and fractional shortening were decreased.It was found that there were statistically significant differences between the degree of plaque and some parameters of cardiac measurement in mice.There were significant differences in left ventricular end-systolic dimension,left ventricular end-diastolic dimension,left ventricular end-systolic volume,left ventricular end-diastolic volume,stroke volume,LVEF,fractional shortening and cardiac output among the groups(P<0.05).Conclusions:There was a significant negative correlation between plaque degree and WSS.It is concluded that low shear stress plays a crucial role in the formation of carotid atherosclerosis in ApoE-/-mice.Moreover,the structure and function of mouse heart can be clearly displayed in utrasonic instrument special for the small animals.And it provides a noninvasive,low-cost and rapid method t
