摘  要:为提高三角褐指藻(Phaeodactylum tricornutum)生产岩藻黄素与中性脂肪酸的能力,本试验研究了不同浓度葡萄糖酸钠对三角褐指藻细胞生长、比生长速率、叶绿素a含量、岩藻黄素产量、中性脂肪酸含量以及相关关键基因表达的影响。结果表明,200 mmol·L^(-1)葡萄糖酸钠可以极显著提高三角褐指藻生物量,相比对照组提升了75%(P<0.01)。葡萄糖酸钠处理三角褐指藻后,第3天比生长速率达最大值。随着葡萄糖酸钠浓度的升高,三角褐指藻中岩藻黄素产量和叶绿素a含量均呈现先升高后降低的趋势,其中岩藻黄素产量的最大值出现在200 mmol·L^(-1)组,而叶绿素a含量最大值出现在100 mmol·L^(-1)组,分别是对照组的1.79和1.67倍,三角褐指藻中性脂肪酸含量与葡萄糖酸钠浓度呈正相关。实时荧光定量PCR(qRT-PCR)分析结果表明,葡萄糖酸钠可以通过促进lcyb和pds基因的表达来促进三角褐指藻岩藻黄素的积累,可以通过促进FAD2与gpat基因的表达提高中性脂肪酸含量。葡萄糖酸钠处理三角褐指藻后会抑制psbA基因的表达,进而降低三角褐指藻光合作用。综上,葡萄糖酸酸钠可以显著提高三角褐指藻生物量与比生长速率,并且可以通过促进关键基因表达来提高岩藻黄素产量和中性脂肪酸含量。本研究结果为三角褐指藻岩藻黄素和油脂的规模化生产和分子机制提供了理论基础。In order to improve the ability of Phaeodactylum tricornutum to produce fucoxanthin and neutral fatty acids,this study investigated the effects of differential concentrations of sodium gluconate on the cell growth,specific growth rate,chlorophyll a content,fucoxanthin production,neutral fatty acids content and related key gene expression of P.tricornutum.The results showed that 200 mmol·L^(-1) sodium gluconate could significantly increase the biomass of P.tricornutum,which was 75%higher than the control group.After treatment with sodium gluconate,the maximum specific growth rate appeared on the 3rd day.With the increase of sodium gluconate concentration,the production of fucoxanthin and chlorophyll a in P.tricornutum showed a trend of increasing first and then decreasing.The maximum value of fucoxanthin production appeared in the 200 mmol·L^(-1) group,while the maximum value of chlorophyll a content appeared in the 100 mmol·L^(-1) group,which were 1.79 and 1.67 times the content in control group respectively,the content of neutral fatty acids in P.tricornutum was positively correlated with the concentration of sodium gluconate.The results of qRT-PCR analysis showed that sodium gluconate could promote the accumulation of fucoxanthin in P.tricornutum by promoting the expression of lcyb and pds gene,and increase the content of neutral fatty acids by promoting the expression of FAD2 and gpat gene.After treatment with sodium gluconate,the expression of psbA gene in P.tricornutum is inhibited,which in turn reduces the photosynthesis of P.tricornutum.Studies have shown that sodium gluconate can significantly increase the biomass and specific growth rate of P.tricornutum,and can increase the content of fucoxanthin,chlorophyll a,and neutral fatty acid by promoting key genes expressions.The results provide a theoretical basis for large-scale production and molecular mechanism of producing fucoxanthin and fatty acids in Phaeodactylum tricornutum.
