摘 要:目的:采用网络药理学分析方法探讨补阳还五汤对腰椎术后疼痛综合征的作用机制。方法:通过检索中药系统药理学数据库与分析平台(TCMSP)和阅读文献筛选补阳还五汤中可被人体吸收的活性成分及靶点,利用Cytoscape 3.9.0软件构建补阳还五汤活性成分-靶点网络图。利用在线人类孟德尔遗传(OMIM)数据库、Disgenet数据库检索“疼痛,术后(Pain,Postoperative)、炎性疼痛(Inflammatory Pain)、下腰痛(Low Back Pain)、骨痛(Bone Pain)”,将获取的疾病基因靶点与补阳还五汤的靶点取交集,预测补阳还五汤治疗腰椎术后疼痛综合征的靶点基因。使用Cytoscape 3.9.0软件构建补阳还五汤治疗腰椎术后疼痛综合征的“活性成分-靶点”网络及蛋白质-蛋白质相互作用网络,利用“微生信”网站进行靶点基因本体论(GO)分析与京都基因与基因组百科全书(KEGG)分析。筛选补阳还五汤治疗腰椎术后疼痛综合征的“活性成分-靶点”的主要活性成分和关键靶点进行分子对接。结果:从TCMSP和文献中筛选并去除重复后符合标准的活性成分70个、靶点292个,通过OMIM数据库、Disgenet数据库共检索到有关腰椎术后疼痛综合征的靶点共609个。将补阳还五汤靶点和腰椎术后疼痛综合征靶点利用Venny网站取交集后得到补阳还五汤治疗腰椎术后疼痛综合征的靶点70个。补阳还五汤治疗腰椎术后疼痛综合征的“活性成分-靶点”网络和靶点蛋白质-蛋白质相互作用显示,槲皮素、β-谷甾醇、木犀草素、黄芩素、山柰酚为补阳还五汤治疗腰椎术后疼痛综合征的主要活性成分,前列腺素内过氧化物酶合成酶2(Prostaglandin-endoperoxidase Synthase 2,PTGS2)、丝氨酸/苏氨酸蛋白激酶1(Akt Serine/Threonine-protein Kinase 1,AKT1)、转化生长因子-β1(Transforming Growth Factor Beta-1,TGFB1)、基质金属蛋白酶9(Matrix Metalloproteinase 9,MMP9)、肿瘤坏死因子(Tumor Necrosis FactObjective:To investigate the mechanism of the Buyang Huanwu decoction(补阳还五汤)in the treatment of failed back surgery syndrome by network pharmacological.Methods:By searching TCMSP and reading literature,the active ingredients that can be effectively absorbed by human body in the Buyang Huanwu decoction and targets were selected,andO Cytoscape 3.9.0 software was used to construct the active ingredient and target network map of the Buyang Huanwu decoction.OMIM database and Disgenet database were used to search postoperative pain,inflammatory pain,low back pain,and bone pain.The obtained target genes of disease were and the targets of the Buyang Huanwu decoction were intersected to predict the target genes of the Buyang Huanwu decoction in the treatment of failed back surgery syndrome.Cytoscape 3.9.0 software was used to construct the active ingredient-target network and protein interaction network of the Buyang Huanwu decoction in the treatment of failed back surgery syndrome,and Weishengxin website was used for GO analysis and KEGG analysis of target genes.The main active components and key targets of active ingredient-target of the Buyang Huanwu decoction for failed back surgery syndrome were screened for molecular docking.Results:A total of 70 active ingredients and 292 targets conforming to the criteria were achieved TCMSP and literature.A total of 609 failed back surgery syndrome targets were retrieved through OMIM database and Disgenet database.After the intersection of the Buyang Huanwu decoction target and failed back surgery syndrome target using Venny website,and 70 targets of the Buyang Huanwu decoction in treating failed back surgery syndrome were obtained.The active component-target network and the target protein interaction network of the Buyang Huanwu decoction for failed back surgery syndrome showed that quercetin,β-sitosterol,luteolin,baicalein and kaempferol were the main active components of the Buyang Huanwu decoction for treating failed back surgery syndrome.PTGS2,AKT1,TGFB1,MMP9 and
