基于靶向PI3K/AKT/mTOR信号通路分析中医药防治腹膜透析相关性腹膜纤维化

摘  要:磷酸肌醇3激酶(phosphoinositol 3 kinase,PI3K)/丝氨酸/苏氨酸蛋白激酶B(protein kinase B,PKB,又称AKT)/哺乳动物雷帕酶素靶蛋白(mammalian target of rapamycin,mTOR)信号通路与腹膜间皮细胞损伤、腹膜间皮细胞上皮-间充质转化密切相关。中医药基于PI3K/AKT/mTOR信号通路能有效抑制腹膜血管生成、腹膜间皮细胞发生间充质转换以及降低壁层腹膜厚度、改善超滤衰竭,从而延缓腹膜纤维化进程。基于中医药干预PI3K/AKT/mTOR信号通路影响腹膜纤维化发生发展的相关研究,发现PI3K蛋白、AKT蛋白、pAKT蛋白、mTOR蛋白为中医药靶向PI3K/AKT/mTOR信号通路防治腹膜纤维化的主要靶点,并分析抑制新生血管生成和腹膜间皮细胞上皮-间充质转化为中医药靶向PI3K/AKT/mTOR信号通路防治腹膜纤维化的主要机理。The phosphoinositol 3-kinase(PI3K)/serine/threonine protein kinase B(PKB,also known as AKT)/mammalian rapamycin target protein(mTOR)signaling pathway is closely related to peritoneal mesothelial cell injury and epithelial mesenchymal transition of peritoneal mesothelial cells.Traditional Chinese medicine,based on the PI3K/AKT/mTOR signaling pathway,can effectively inhibit peritoneal angiogenesis,mesenchymal transition of peritoneal mesothelial cells,reduce parietal peritoneal thickness,improve ultrafiltration failure,and thus delay the process of peritoneal fibrosis.This article explores the effects of traditional Chinese medicine intervention in the PI3K/AKT/mTOR signaling pathway on the occurrence and development of peritoneal fibrosis.It is found that PI3K protein,AKT protein,pAKT protein and mTOR protein are the main targets of traditional Chinese medicine targeting the PI3K/AKT/mTOR signaling pathway to prevent and treat peritoneal fibrosis.Besides,the main mechanism of inhibiting neovascularization and epithelial mesenchymal transition of peritoneal mesothelial cells is analyzed as a traditional Chinese medicine targeted PI3K/AKT/mTOR signaling pathway for preventing and treating peritoneal fibrosis.
