亲水粘附型壳聚糖织物的制备及其止血研究

摘  要:通过吸收和浓缩血液,实现对红细胞和血小板的快速聚集,对止血尤为重要。为了提高壳聚糖织物对血液的吸收效果,采用丙烯酸对壳聚糖纤维进行亲水性改性,得到不同羧乙基取代度的羧乙基化壳聚糖(CECS)纤维;为了提高壳聚糖织物的组织粘附性能,采用HCl对壳聚糖纤维进行氨基质子化处理,得到水溶型质子化壳聚糖(CS-HCl)纤维。进一步,以聚乳酸(PLA)长丝为芯层、CECS纤维为鞘层,通过环锭纺纱工艺,得到PLA@CECS包芯纱线,再以PLA@CECS包芯纱线作为芯层、CS-HCl纤维为鞘层,通过摩擦纺工艺,得到具有三层结构的PLA@CECS@CS-HCl包芯纱线。继而经针织加工,得到PLA@CECS@CSHCl织物(壳聚糖织物)。壳聚糖织物表面具有良好的亲水性,也显示出良好的血液垂直吸收性。同时,溶胀的壳聚糖织物也具有优异的血液封堵能力。壳聚糖织物能紧密粘附在组织表面。在兔肝脏和兔股动脉损伤模型中,高羧乙基取代度的壳聚糖织物F-H-fabric的止血时间和出血量均显著少于纱布,显示其优秀的止血性能。此外,壳聚糖织物具有良好的细胞相容性和血液相容性。壳聚糖织物未来在致命性大出血急救领域具有广阔的应用前景。To achieve rapid aggregation of red blood cells and platelets by absorbing and concentrating blood is particularly important for hemostasis.In order to improve the blood absorption capacity of chitosan fabric,carboxyethyl chitosan(CECS)fiber with different degree of carboxyethyl substitution was synthesized by the reaction between acrylic acid and chitosan fiber.Meanwhile,to improve the tissue adhesion of chitosan fab-ric,water-soluble protonated chitosan fiber(CS-HCl)was prepared by modifying chitosan fiber with HCl.Furthermore,polylactic acid(PLA)@CECS core-spun yarns were obtained by ring spinning of(PLA)filament as core layer and CECS fiber as sheath layer,then PLA@CECS@CS-HCl core-spun yarns with three-layer structure was obtained by covering CS-HCl fiber on PLA@CECS core-spun yarns.Finally,PLA@CECS@CS HCl fabric(chitosan fabric)was prepared by knitting.Chitosan fabric has good hydrophilicity and also shows good vertical absorption of blood.Meanwhile,swollen chitosan fabrics also have excellent blood blocking property.Chitosan fabrics can tightly adhere to the surface of tissues.In the rabbit liver model and femoral ar-tery injury model,the hemostatic time and blood loss of the fabric(e.g.F-H-fabric)were significantly less than those of gauze,showing its excellent hemostatic properties.In addition,the fabric had good cytocompati-bility and blood compatibility.Chitosan fabric is a promising hemostatic material for fatal massive hemor-rhage in the future.
