摘　　要：目的探讨槲皮素(Quercetin)对小鼠压力负荷性心力衰竭(HF)的影响及作用机制。方法30只SPF级雄性C57BL/6小鼠随机分成假手术组(Sham)、心力衰竭组(HF)和HF+Quercetin组,采用主动脉弓缩窄术(TAC)构建小鼠压力负荷性HF,术后腹腔注射给予Quercetin,2次/日,连续6周。小动物超声检测小鼠心功能;称量心脏重量并计算心脏重量指数;HE和Masson染色观察小鼠心脏组织病理学改变和纤维化程度;Western blot检测小鼠心肌组织Bax、Bcl-2、cleaved caspase-3、Wnt3a和β-catenin蛋白的表达。结果Quercetin显著降低HF小鼠心脏重量和心脏体重比。超声心动图显示Quercetin显著改善HF小鼠的心功能,并降低升主动脉内径。Quercetin治疗改善HF小鼠心脏病理改变和纤维化程度;降低HF小鼠心脏组织Bax、cleaved caspase-3、Wnt3a和β-catenin蛋白的表达,升高Bcl-2的表达。结论Quercetin通过抑制Wnt3a/βcatenin信号通路对HF小鼠心肌提供保护作用。Objective To investigate the effect and mechanism of Quercetin on stress load heart failure(HF)in mice.Methods Thirty SPF grade male C57BL/6 mice were randomly divided into sham group,HF group with heart failure and HF+Quercetin group.Pressure loaded HF mice were constructed by aortic arch reduction(TAC)and Quercetin mice were injected intraperitoneally twice a day for 6 weeks.The heart function of mice was detected by ultrasound in small animals.The heart was weighed and calculate the heart weight index.HE and Masson staining were used to observe cardiac histopathological changes and fibrosis degree.The expressions of Bax,Bcl-2,cleaved caspase-3,Wnt3a and β-catenin in mouse myocardial tissue were detected by Western blot.Results Quercetin significantly reduced heart weight and heart weight ratio in HF mice.Echocardiography showed that Quercetin significantly improved cardiac function and reduced the ascending aorta diameter in HF mice.Quercetin treatment improved cardiac pathological changes and fibrosis degree in HF mice.The expressions of Bax,cleaved caspase-3,Wnt3a andβ-catenin were decreased in HF mice,and the expression of Bcl-2 was increased.Conclusion Quercetin protects the myocardium of HF mice by inhibiting the Wnt3a/βcatenin signaling pathway.
