摘  要:目的探讨抵抗素(Resistin)在体外人脑微血管内皮细胞(HBMEC)血管生成过程中的作用。方法血管生成实验比较Resistin处理组以及对照组HBMECs的血管生成情况。增殖实验和划痕迁移实验检测细胞增殖和细胞迁移状况。应用β-半乳糖苷酶染色检测细胞衰老状况。应用四甲基罗丹明乙酯(TMRM)线粒体染色后观察HBMECs的线粒体膜电位状况以及线粒体活性。PCR方法检测HBMECs的线粒体DNA含量的变化。结果Resistin抑制体外培养HBMECs的血管生成,诱导HBMECs衰老,降低线粒体活性但并不影响HBMECs中线粒体DNA的含量,对HBMECs增殖及迁移无影响。结论Resisitn抑制脑微血管生成与下调线粒体活性相关。Objective To investigate the role of Resistin in the angiogenesis of human brain microvascular endothelial cell(HBMEC)in vitro.Methods The angiogenesis of HBMECs was studied between Resistin-treated group and the normal control group.The proliferation and migration of HBMECs were determined by proliferation test and wound healing test respectively.β-galactosidase staining was used to study the senescence of HBMECs.The mitochondrial activity of HBMECs were observed by TMRM(Tetramethylrhodamine methyl ester)staining.PCR was used to detect mitochondrial DNA content of HBMECs.Resulsts Resistin inhibited the angiogenesis of HBMECs in vitro,induced the senescence of HBMECs,reduced the activity of mitochondria but did not affect the content of mitochondrial DNA in HBMECs,not affect the proliferation and migration of HBMECs.Conclusion The inhibition of Resistin on brain angiogenesis is related to down regulating mitochondrial activity.
