摘 要:目的研究当归多糖对再生障碍性贫血小鼠外周血细胞和细胞因子表达水平的调控作用。方法60只ICR雄性小鼠随机分为对照组、模型组、当归多糖低剂量(100 mg/kg)组和高剂量(200 mg/kg)组。收集小鼠外周和骨髓来源的血液样本,检测小鼠外周血细胞的数量和骨髓单个核细胞数量。酶联免疫吸附法检测小鼠血清中IL-2、IL-4、IFN-γ、TNF-α、T-AOC和MDA等细胞因子水平。qPCR和Western blot方法检测脾脏组织中Notch1和DLL4的mRNA和蛋白水平的表达。结果与对照组相比,模型组小鼠血清中炎症细胞因子表达升高,当归多糖治疗组能有效降低IL-2、IFN-γ和TNF-α表达水平,但是IL-4没有统计学差异。与对照组相比,模型组小鼠脾脏组织中T-AOC和MDA表达升高,Notch1的表达水平降低,DLL4的表达水平升高;药物治疗后,小鼠脾脏组织中T-AOC和MDA表达降低,Notch1的表达水平升高,DLL4的表达水平降低。结论当归多糖降低再生障碍性贫血小鼠炎性细胞因子的表达水平,减少氧化应激损伤。Objective To investigate the regulation of angelica polysaccharide on peripheral blood cell and cytokine expression level in aplastic anemia mice.Methods Sixty ICR male mice were randomly divided into control group,model group,angelica polysaccharide low dose(100 mg/kg)group and high dose(200 mg/kg)group.Peripheral blood samples and bone marrow derived blood samples were collected to detect the number of peripheral blood cells and bone marrow mononuclear cells in mice.Levels of IL-2,IL-4,IFN-γ,TNF-α,T-AOC and MDA in serum were measured by ELISA.The mRNA and protein levels of Notchl and DLL4 in spleen tissues were detected by qPCR and Western blot.Results Compared with the control group,the expressions of inflammatory cytokines were increased in the model group,and the levels of IL-2,IFN-γ and TNF-α were effectively reduced in the angelica polysaccharide treatment group,but there were no statistically significant differences in IL-4.Compared with the control group,the expressions of T-AOC and MDA in the spleen of the model group were increased,the expression of Notch1 was decreased and the expressions of DLL4 were increased.After drug treatment,the expressions of T-AOC and MDA were decreased,the expression of Notch1 was increased and the expression of DLL4 was decreased.Conclusion Angelica polysaccharide could decreased the expression level of inflammatory cytokines and oxidative stress injury in aplastic anemia mice.
