中乌宁对庆大霉素所致急性肾损伤大鼠的保护作用研究

摘  要:目的:观察不同剂量中乌宁对庆大霉素(GM)诱导急性肾损伤(AKI)大鼠肾功能改善作用,探讨其可能的作用机制。方法:腹腔注射GM 10 d以建立AKI模型后,连续给予中乌宁(2 mg·kg^(-1)及4 mg·kg^(-1))15 d;观察体重、肾脏指数、尿量及尿微量白蛋白(mALB)、血清中肌酐(CREA)、尿素氮(BUN)水平;采用ELISA法检测大鼠血清及肾组织中肾损伤分子-1(KIM-1)、中性粒细胞明胶酶相关脂质运载蛋白(NGAL)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-18(IL-18)水平。结果:与模型组相比,中乌宁2 mg·kg^(-1)组大鼠肾脏指数、尿mALB明显降低(p<0.05,p<0.01)。中乌宁4 mg·kg^(-1)组大鼠血清中BUN明显降低(p<0.05)。中乌宁2 mg·kg^(-1)及4 mg·kg^(-1)组大鼠血清KIM-1、NGAL含量显著减少(p<0.01)。中乌宁2 mg·kg^(-1)组大鼠血清TNF-α、IL-1β及IL-18水平降低,中乌宁4 mg·kg^(-1)组TNF-α及IL-18水平降低(p<0.05,p<0.001)。结论:中乌宁可改善GM诱导AKI大鼠的急性肾损伤,其机制可能与下调炎症因子TNF-α、IL-1β及IL-18水平有关。Objective:To observe the effects of different doses of mesaconine on the improvement of renal function in rats with gentamicin(GM)-induced acute kidney injury(AKI)and to investigate the mechanism.Method:After intraperitoneal injection of GM for 10 d to establish AKI models,mesaconine(2 mg·kg^(-1)and 4 mg·kg^(-1))was given continuously for 15 d.Body weight,renal index,urine volume and urine microalbumin(mALB),serum creatinine(CREA)and blood urea nitrogen(BUN)levels were observed.ELISA was used to detect renal injury molecule-1(KIM-1),neutrophil gelatinase-associated lipid transport protein(NGAL),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-18(IL-18)levels in serum or kidneys of rats.Result:Compared with the model group,the renal index and urinary mALB were signiffcantly lower in mesaconine 2 mg·kg^(-1)group(p<0.05,p<0.01).The serum BUN was signiffcantly reduced in mesaconine 4 mg·kg^(-1)group(p<0.05).The levels of KIM-1and NGAL were signiffcantly reduced in the mesaconine 2 mg·kg^(-1) and 4 mg·kg^(-1)groups(p<0.01).TNF-α,IL-1βand IL-18 levels were reduced in mesaconine 2 mg·kg^(-1)group,and TNF-αand IL-18 levels were reduced in mesaconine 4 mg·kg^(-1)group(p<0.05,p<0.001).Conclusion:Mesaconine can improve the renal function of GM-induced AKI rats,and the mechanism may be related to the down-regulation of inflammatory factors TNF-α,IL-1βand IL-18 levels.
