摘  要:目的探索SD大鼠孕期六氟环氧丙烷二聚羧酸(HFPO-DA)暴露对雄性子代体质量、睾丸组织及睾酮分泌的影响。方法建立大鼠染毒模型,将妊娠0.5 d的SD大鼠随机分成4组,分别在整个妊娠期间(妊娠0.5~22 d)口服去离子水,低(1 mg/kg)、中(10 mg/kg)、高(100 mg/kg)剂量组的HFPO-DA。在孕鼠完成分娩(妊娠22 d)后,在生后第1天时,记录其雄性子鼠的体质量变化,采用H-E染色观察子代睾丸组织的病理学形态。在生后第21天时,记录其雄性子鼠的体质量及睾丸质量,采用H-E染色观察其睾丸组织的病理学形态,同时测量睾丸组织的曲细精管内径,采用ELISA法检测子鼠血清中的睾酮水平。结果较对照组而言,在生后第1天时,各个剂量组的子鼠体质量偏小,且具有显著的统计学差异(P<0.05);在光学显微镜下,最高剂量组的子鼠睾丸组织变小,曲细精管内的精原细胞处于原始核分裂像。在生后第21天时,较对照组而言,暴露在中、高剂量组的子鼠体质量及睾丸质量呈下降趋势,但各剂量组之间的睾丸/体质量的比值没有统计学差异(P>0.05);各剂量组之间睾丸组织病理学形态未见明显异常,同时睾丸组织曲细精管管腔内径未见差异(P>0.05);各剂量组之间子代的睾酮水平也没有统计学差异(P>0.05)。结论孕期HFPO-DA暴露可以使刚出生的雄性子代体质量下降及抑制其睾丸精原细胞的发育,但随着子代的生长发育,到断乳期(生后第21天)时,其对体质量的抑制作用减弱,而对睾丸精原细胞发育的抑制作用消失,同时对曲细精管内径及睾酮分泌无任何影响。Objective To explore the effects of HFPO-DA exposure during gestation in SD rats on body weight,testicular tissues and testosterone secretion of male offspring.Methods A exposure toxicity model of rat was established,rats at gestation days 0.5(GD0.5)were randomly divided into 4 groups and orally administered HFPO-DA in deionized water,low(1 mg/kg),medium(10 mg/kg)and high(100 mg/kg)dose groups during the gestation period(GD0.5-GD22),separately.After the pregnant rats completed delivery(GD22),at postnatal day 1(PND1),the body weight of male pups were observed and the histopathological morphology of their testicular tissues were observed by H-E staining.At postnatal day 21(PND21),the body weight and testicular weight of the male offspring were recorded,and the histopathological morphology of the testes were observed by H-E staining.Meanwhile,the inner diameter of seminiferous tubules were measured,and the serum testosterone level of the offspring was measured by ELISA.Results At postnatal day 1(PND1),the body weights of male pups were smaller and significant statistically different(P<0.05)in each dose group compared with the control group,while under light microscopy,testes in the highest dose group became smaller in size and spermatogonia in the varicocele were in primitive nuclear division images compared with the control group.At postnatal day 21(PND21),the body weight and the weight of offspring's testes exposed to medium and high dose tended to decrease compared with the control group,but the testis/body weight ratio was not statistically different between the dose groups(P>0.05).No significant abnormalities in the histopathological morphology of the testes were observed in all dose groups,meanwhile,there was no difference in the inner diameter of the seminiferous tubules in each dose group(P>0.05).There were also no statistically significant differences in testosterone levels between the dose groups(P>0.05).Conclusion Exposure to HFPO-DA during gestation resulted in reducing the body weight of the male pups a
