摘 要:目的:建立了俄色叶(变叶海棠)配方颗粒的最佳提取工艺条件。方法:采用高效液相色谱法同时测定俄色叶(变叶海棠)中的绿原酸、金丝桃苷、异槲皮苷、槲皮苷、根皮苷、槲皮素、根皮素的含量。在单因素试验的基础上,以浸泡时间、料液比、提取时间、提取次数为考察因素,上述7种成分含量、总黄酮、出膏率的综合评分为考察指标,采用Box-Behnken响应面法优化俄色叶(变叶海棠)配方颗粒的提取工艺。结果:俄色叶(变叶海棠)配方颗粒的最佳提取工艺为俄色叶饮片加水煎煮2次,第一次加15倍量水,浸泡30 min,煎煮45 min;第二次加13倍量水,煎煮45 min,滤过,合并煎液,即得。经过3次实验验证,所得平均综合评分为0.91(RSD=0%,n=3),与预测综合评分(0.938)的相对误差较小(3.08%)结论:Box-Behnken响应面法结合多指标综合评分法可用于俄色叶(变叶海棠)配方颗粒提取工艺优化,且所得最佳提取工艺稳定、简单、重现性好,可为俄色叶(变叶海棠)配方颗粒的制备及工业化大生产提供基础。Objective:The optimum extraction conditions of the formula granules of Malus toringoides(Rehd.)Hughes were established.Methods:The contents of chlorogenic acid,hypericin,isoquercetin,quercetin,rhizoside,quercetin and phloretin in the leaves of E′seye were determined by HPLC.Based on single factor test,with soaking time,solid-liquid ratio,extraction time and extraction times as factors,using comprehensive scores of the contents of above seven components,total flavonoids,paste yield as indexes.the extraction technology of Malus toringoides(Rehd.)Hughes formulation granules was optimized by Box-Behnken response surface methodology.Results:The optimal extraction technology was to add water to decoction pieces of E′seye for two times,the first time adding 15 times the amount of water,soaking for 30 min,decocting for 45 min;Add 13 times the amount of water for the second time,fry for 45 min,strain,combine the decoction,and get.After 3 times of validation tests,the average comprehensive score was 0.91(RSD=0%,n=3),relative error of which with predicted score(0.938)was 3.08%.Conclusion:Box-Behnken response surface methodology combined with multi-index comprehensive score can be used for the extraction technology of Malus toringoides(Rehd.)Hughes formulation granules,and the optimized extraction technology is stable,feasible and reproducible,which could provide a basis for the preparation and industrial production of Malus toringoides(Rehd.)Hughes formula granules.
