摘  要:目的:探讨补肾通络方对膝骨关节炎大鼠软骨损伤、氧化应激及TGF-β/SMAD2信号通路的作用机制。方法:45只大鼠按照随机数字表法分为正常组、模型组、补肾通络低剂量组、补肾通络高剂量组、西药组5组,每组9只,除正常组外,其余组均建立膝骨关节炎模型,补肾通络低、高剂量组分别灌胃补肾通络颗粒制成的混悬液(1.15g/kg)、(4.65g/kg),西药组灌胃美洛昔康溶液(0.073g/kg),其余大鼠灌胃等量蒸馏水。HE染色观察各组大鼠膝关节软骨形态;分析天平观察各组大鼠软骨含水率;ELISA法检测各组大鼠关节液MDA、SOD含量;PCR测定各组大鼠软骨中IGF-1、BMP-7、OPN水平;免疫印迹检查各组大鼠TGF-β1、Smad2、Smad7蛋白表达。结果:正常组软骨含水率为(41.35±1.10)%,模型组软骨含水率为(52.41±5.22)%,低、高剂量补肾通络组软骨含水率分别为(49.18±3.15)%、(42.38±1.14)%,西药组软骨含水率为(43.13±1.15)%。与正常组相比,模型组软骨含水率升高(P<0.05),与模型组相比,低、高剂量补肾通络组软骨含水率降低(P<0.05),低、高剂量补肾通络组软骨含水率逐渐降低(P<0.05);正常组软骨表面光滑,细胞排列规律,间质均匀,潮线清晰,模型组软骨面糜烂,细胞排列无规则,存在间质纤维化,无潮线,低、高剂量补肾通络组及西药组软骨形态改善,补肾通络高剂量组尤其明显;与正常组相比,模型组大鼠MDA、OPN、TGF-β1、Smad2升高,SOD、IGF-1、BMP-7、Smad7降低(P<0.05),与模型组相比,低、高剂量补肾通络组MDA、OPN、TGF-β1、Smad2降低,SOD、IGF-1、BMP-7、Smad7升高(P<0.05);西药组与补肾通络高剂量组比较软骨含水率、MDA、SOD、IGF-1、BMP-7、OPN、TGF-β1、Smad2及Smad7无统计学差异(P>0.05)。结论:补肾通络方可改善软骨损伤形态,减少软骨含水率,降低氧化应激损伤,抑制炎症反应,其作用机制与调控TGF-β/SMAD2通路的表达相关。Objective:To explore the effects of Bushen Tongluo Formula on cartilage damage,oxidative stress,and TGF in knee osteoarthritis rats-β/The mechanism of action of the SMAD2 signaling pathway.Methods:Forty five rats were randomLy divided into five groups:normal group,model group,low dose group of Bushen Tongluo,high dose group of Bushen Tongluo,and western medicine group,with 9 rats in each group.Except the normal group,the rest groups were established with knee osteoarthritis models.The low and high dose groups of Bushen Tongluo were respectively given the suspension(1.15g/kg)and(4.65g/kg)made of Bushen Tongluo granules,and the western medicine group was given meloxicam solution(0.073 g/kg),The remaining rats were given an equal amount of distilled water by gavage.HE staining was used to observe the morphology of knee joint cartilage in each group of rats;Analytical balance was used to observe the cartilage moisture content of rats in each group;ELISA method was used to detect the content of MDA and SOD in the joint fluid of rats in each group;PCR was used to determine the levels of IGF-1,BMP-7,and OPN in the cartilage of rats in each group;Immunoblotting examination of TGF in rats of each group-β1.Smad2 and Smad7 protein expression.Result:The cartilage moisture content of the normal group was(41.35±1.10)%,while that of the model group was(52.41±5.22)%.The cartilage moisture content of the low and high dose kidney tonifying and meridian dredging groups was(49.18±3.15)%and(42.38±1.14)%,respectively.The cartilage moisture content of the Western medicine group was(43.13±1.15)%.Compared with the normal group,the model group had an increase in cartilage moisture content(P<0.05).Compared with the model group,the low and high doses of tonifying the kidney and unblocking the collaterals group had a decrease in cartilage moisture content(P<0.05),while the low and high doses of tonifying the kidney and unblocking the collaterals group gradually decreased cartilage moisture content(P<0.05);The normal group had smooth c
