摘  要:目的探讨姜黄素通过调控miR-134发挥神经保护及潜在抗癫痫作用的机制。方法按照随机数字表法将48只大鼠分为对照组、癫痫组和姜黄素组3组,每组16只。除对照组外,其余两组采用氯化锂-匹罗卡品诱导制备癫痫持续状态大鼠模型。造模后第2天开始,姜黄素组大鼠每天固定时间腹腔注射2 mL/kg姜黄素,而癫痫组和对照组每天固定时间腹腔注射等量二甲基亚砜溶液,持续10 d。在造模后第14天,通过颈椎脱臼法处死3组大鼠,采用Timm染色观察大鼠海马组织中苔藓纤维出芽程度,qRT-PCR法检测大鼠海马组织及血清中miR-134表达水平,Western blot法检测大鼠海马组织中LIM激酶1(LIMK1)蛋白表达水平。结果与对照组比较,癫痫组大鼠齿状回和CA3区苔藓纤维出芽程度评分均升高(均P<0.05);与癫痫组比较,姜黄素组大鼠齿状回和CA3区苔藓纤维出芽程度评分均降低(均P<0.05)。与对照组比较,癫痫组大鼠海马组织中miR-134表达水平升高(P<0.05),LIMK1蛋白表达水平下降(P<0.05);与癫痫组比较,姜黄素组大鼠海马组织中miR-134表达水平下降(P<0.05),LIMK1蛋白表达水平升高(P<0.05)。与对照组比较,癫痫组和姜黄素组大鼠血清中miR-134表达水平均下降(均P<0.05)。结论姜黄素可抑制癫痫持续状态大鼠海马组织中miR-134表达,上调LIMK1蛋白表达水平,影响突触重塑,可能因此发挥神经保护及潜在的抗癫痫作用。Objective To investigate the mechanism of curcumin exerting neuroprotective and potential antiepileptic effects through modulation of miR-134.Methods Forty-eight rats were divided into three groups of control,epilepsy and curcumin according to the random number table method,with 16 rats in each group.Except for the control group,the remaining two groups applied lithium chloride-pilocarpine to induce rat models of status epilepticus.After modeling,rats in the curcumin group were intraperitoneally injected with 2 mL/kg of curcumin,while rats in the epilepsy group and the control group were intraperitoneally injected with 2 mL/kg of dimethylsulfoxide,both at a fixed time every day from day 2,and the drugs were administered continuously for 10 d.Rats were sacrificed by cervical dislocation on the 14th day after successful modeling,and the degree of mossy fiber sprouting in the hippocampal tissue of rats was observed by Timm staining.The expression levels of miR-134 in rat hippocampal tissue and serum were detected by qRT-PCR,and the protein expression level of LIM kinase 1(LIMK1)in rat hippocampal tissue was detected by Western blot.Results Compared with the control group,the scores of the degree of mossy fiber outgrowth in the dentate gyrus and CA3 region of rats in the epilepsy group were both elevated(both P<0.05);while compared with the epilepsy group,those scores in the curcumin group were decreased(both P<0.05).Compared with the control group,the expression level of miR-134 in the hippocampal tissue of rats in the epilepsy group was increased(P<0.05),while the expression level of LIMK1 protein was decreased(P<0.05);Compared with the epilepsy group,the miR-134 level in the hippocampal tissue of rats in the curcumin group was decreased(P<0.05),while LIMK1 protein expression level was increased(P<0.05).Compared with the control group,serum miR-134 expression levels of rats in the epilepsy and curcumin groups were both decreased(both P<0.05).Conclusion Curcumin inhibits miR-134 expression and upregulates LIMK1 leve
