摘  要:昆虫是危害农林作物的主要害虫,利用微生物防治有害昆虫是减少农林作物损失的有效途径之一。植物内生菌是生防菌的重要来源。以家蚕蛹外皮几丁质为筛选培养基主要成分,从西藏垫状雪灵芝(Arenaria pulvinata Edgew.)内生菌中筛选产高活性几丁质酶的菌株。透明圈法与DNS法酶活性测定相结合,筛选出一株降解几丁质能力较强、酶活性较高的菌株ZLB-158,根据形态学特征与16S rRNA基因序列鉴定,初步将该菌株归属为侧孢短芽孢杆菌(Brevibacillus laterosporus)。该菌株在蚕蛹胶体几丁质平皿上培养48 h形成的降解圈与菌落直径比为2.6,初始发酵条件下酶活力为0.404 U/mL。ZLB-158菌株产几丁质酶活性较高,在昆虫害虫防治方面具有潜在应用价值。Insects are the major pests that harm agroforestry crops. One of the effective ways to reduce the economic loss is microbial control method. Plant endophytes are important source of biocontrol agents. In this study, a bacterial strain ZLB-158 with strong chitin degradation ability and high chitinase activity was screened from the endophytes of Arenaria pulvinata Edgew. of Xizang with chitin from the outer skin of silkworm pupa being the main component of the screening medium, and by combining the transparent circle method and DNS method for enzyme activity determination. Based on morphological characteristics and 16S rRNA gene sequencing, the strain was preliminarily classified as Brevibacillus laterosporus. The ratio of the transparent circle to colony diameter was 2.6 after the colonies were cultured on chitin agar plates for 48 h. Under the initial fermentation conditions, the chitinase activity was 0.404 U/mL. ZLB-158 exhibited high chitinase activity and has potential application value in the control of insect pests.
