摘 要:为了明确根结线虫种类,采用形态学和分子生物学方法,对甘肃省临洮县胡萝卜上的根结线虫进行鉴定,并测定其致病性。结果表明,该群体雌虫虫体梨形或柠檬形,乳白色,口针粗壮,中食道球近圆形。雌虫会阴花纹呈卵圆形,背弓较平缓,肛门处有刻点。雄虫虫体细长,尾部钝圆,交合刺针状,成对。2龄幼虫虫体细长,呈蠕虫形,唇区无明显缢缩,口针发达,口针基部球小而圆,尾部透明区明显,尾尖钝圆。利用特异性引物Mh-R/Mh-F扩增得到大小为462 bp的特异性片段,与北方根结线虫(Meloidogyne hapla)一致。基于根结线虫属rDNA ITS和28S-rDNA D2D3区序列构建系统发育树,发现该群体均与北方根结线虫序列聚为一支。将该线虫接种胡萝卜,其症状与田间发病症状一致,35 d后分离线虫,其形态特征与北方根结线虫相符。综上,形态学特征结合分子生物学方法,将该病原鉴定为北方根结线虫(M.hapla),这是甘肃省首次报道北方根结线虫危害胡萝卜类蔬菜作物。Morphological and molecular biological methods were used to identified the nematode popu-lation affecting carrots in Lintao county,Gansu province.The results showed that the female nema-todes exhibiting pear-shaped or lemony bodied,were milky white with a robust stylet,and had round median bulbs.The perineal patterns of females were oval with a low dorsal arch and punctations,and typically located above the anus.The male nematodes displayed elongated bodies with obtuse tails and a pair of acicular spicula.The second-stage juveniles(J2)had elongated and helminth-shaped bodies with no obvious constriction in the labial area.Their stylets were robust with small and round basal balls,and their tails were obtuse with obvious transparent areas.A specific 462 bp fragment was am-plified from the population using Mh-R/Mh-F specific primers,consistent with that of Meloidogyne hapla.Phylogenetic trees constructed based on the rDNA ITS and 28S-rDNA D2D3 sequences of the genus meloidogyne,indicated that this population clustered with the sequences of M.hapla.This nem-atode population inoculated on carrots led to the symptoms consistent with those of the diseased ones in the field.After 35 days,the nematode was isolated,and its morphological characteristics matched those of M.hapla.In conclusion,both morphological characteristics and molecular biological methods identify the the pathogen as M.hapla.This is the first report of M.hapla infecting carrot vegetable crop in Gansu province.
