摘  要:目的:大鼠成骨细胞膜片的体外构建,通过形态学检测及增殖活性检测,评估其活性。方法:原代培养大鼠成骨细胞,纯化鉴定后体外构建成骨细胞膜片,用扫描电镜观察、组织学形态学检查,并测定厚度变化及细胞增殖情况。结果:经过5d培养的成骨细胞连接成片,刮刀可刮下。HE染色见丰富的细胞外基质。扫描电镜观察到细胞层间有间隙孔。与正常培养的成骨细胞相比,细胞膜片第1周至第2周增殖迅速,第2周增殖活力最高。随着培养时间的延长,厚度增加,厚度在1周至2周显著增加,对照组直到3周才形成膜片,且厚度低于维生素C组。结论:维生素C促成骨细胞增殖形成细胞膜片,本方法可以稳定体外构建成骨细胞膜片,2周内膜片厚度增加显著并且增殖活力较好。Objective To construct osteoblast membranes from rat cells in vitro and evaluate their activity through morphological and proliferation assays.Methods Primary cultures of rat osteoblasts were established,purified,and used to construct osteoblast membranes in vitro.The membranes were observed via scanning electron microscopy and subjected to histological morphological examinations.Changes in thickness and cell proliferation were measured.Results Osteoblasts cultured for 5 days formed interconnected membranes detachable by scraping.Hematoxylin and eosin staining revealed abundant extracellular matrix.Scanning electron microscopy showed intercellular gaps.Compared to normally cultured osteoblasts,the proliferation of membrane cells was most rapid between the first and second weeks,peaking in the second week.Thickness notably increased with prolonged culture,significantly rising between the first and second weeks.The control group formed membranes only by the third week,with thickness lower than the Vitamin C group.Conclusions Vitamin C promotes osteoblast proliferation and the formation of cell membranes.This method enables stable in vitro construction of osteoblast membranes,with significant thickness increase within two weeks and good proliferation activity.
