摘 要:目的 研究江西产的龙胆科獐牙菜属植物狭叶獐牙菜90%乙醇提取物的乙酸乙酯萃取部分的化学成分。方法选取江西产的狭叶獐牙菜,用90%的乙醇提取后,依次以石油醚、二氯甲烷、乙酸乙酯、正丁醇萃取,得到各部位的萃取物浸膏。对各部位进行体外的HepG2细胞葡萄糖消耗实验筛选出活性部分,对活性部分进行化学成分研究。结果 体外实验显示,乙酸乙酯萃取部分较好。采用各种柱色谱法对乙酸乙酯部分进行化学成分分离,得到化合物15个,全部为已知化合物,其中山酮及其苷12个,四氢山酮苷1个,环烯醚萜2个。通过波谱数据分析(^(1)HNMR,^(13)CNMR)和文献对照鉴定了化合物的结构,其中化合物1~15为首次从狭叶獐牙菜中分离得到。结论 狭叶獐牙菜乙酸乙酯部分增加细胞葡萄糖消耗的作用可能与其富含的山酮及其苷类成分有关。Objective To study the chemical constituents of ethyl acetate extract from 90% ethanol extract of Swertia angustifolia Buch.-Ham,a plant of Gentianaceae from Jiangxi.Methods Swertia angustifolia Buch.-Ham from Jiangxi Province was extracted with 90% ethanol,followed by Petroleum ether,dichloromethane,ethyl acetate,and n-butanol to obtain the extracts of each part.The active part was screened out by glucose consumption test of HepG2 cells,and then the chemical composition of the active part was studied.Results The in vitro experiment showed that the ethyl acetate extraction was better.The chemical constituents of ethyl acetate were separated by various Column chromatography,through the chemical separation of S.angustifolia,15 compounds were obtained,all of which are known compounds,including 12 xanthones and xanthone glycosides,1 tetrahydro xanthone glycosides and 2 iridoids.Through spectral data analysis(^(1)HNMR,^(13)CNMR)and literature comparison,their molecular structure were characterized.Conclusion The effect of ethyl acetate of Swertia angustifolia on increasing glucose consumption in cells may be related to the xanthone and its glycosides.
