摘 要:【目的】分析水稻(Oryza sativa)小分子热激蛋白(Small heat shock protein,sHSP)基因Os02g0782500对逆境胁迫和激素的响应模式,为进一步研究Os02g0782500在逆境胁迫和激素响应过程中的功能提供理论依据。【方法】在水稻‘中花11’中克隆获得Os02g0782500,并对其进行生物信息学分析;同时利用定量PCR(qRT-PCR)技术分析Os02g0782500在水稻不同组织及不同激素和非生物胁迫处理下的表达模式。【结果】Os02g0782500编码区全长为519 bp,编码一个含有HSP20保守结构域的s HSP。系统进化分析显示,Os02g0782500与玉米等单子叶植物中的同源蛋白亲缘关系较近。顺式作用元件分析表明,Os02g0782500的启动子区含有28个植物激素响应元件和35个环境胁迫响应元件。qRT-PCR分析表明,Os02g0782500在水稻叶片中的表达量最高;且Os02g0782500的表达受6-BA、GA3、IAA、高温、低温、PEG6000和NaCl的诱导,推测其在水稻非生物胁迫响应过程中具有重要作用。【结论】明确了Os02g0782500的表达受高温、低温、6-BA、GA3和IAA等外界因素调控,推测Os02g0782500可能通过IAA等激素信号转导途径参与水稻的非生物胁迫响应。【Objective】Analysis of the response pattern of small heat shock protein(sHSP)gene Os02g0782500 to stress and hormones in rice(Oryza sativa)could provide theoretical basis for further research on the functions of Os02g0782500 in stress and hormone responses.【Method】In this study,Os02g0782500 was cloned from‘Zhonghua 11’and bioinformatic analysis was performed.Quantitative real-time PCR(qRT-PCR)was used to analyze the expression patterns of Os02g0782500 in different tissues and under different hormones and abiotic stress treatments.【Result】The gene Os02g0782500 had a coding region with a length of 519 bp,which encoded a small heat shock protein containing one conserved HSP20 domain.The phylogenetic analysis showed that Os02g0782500 was closely related to homologous proteins in maize and other monocotyledonous plants.The analysis of cis-acting elements showed that the promoter region of Os02g0782500 contained 28 plant hormone response cis-elements and 35 environmental stress response cis-elements.The qRT-PCR analysis showed that Os02g0782500 had the highest expression level in rice leaves.And the expression level of Os02g0782500 was induced by 6-BA,GA3,IAA,high temperature,low temperature,PEG6000 and NaCl,which suggested that OS02G0782500 was essential for the abiotic stress responses in rice.【Conclusion】This study clarified that the expression of Os02g0782500 was regulated by external factors such as high temperature,low temperature,6-BA,GA3 and IAA,which provided a foundation for the subsequent study of the biological function of Os02g0782500.It is suggested that Os02g0782500 may be involved in abiotic stress response of rice through IAA and other hormone signal transduction pathways.
