白藜芦醇对鸭卵巢组织冻融后移植效果的影响

摘  要:为探究不同浓度白藜芦醇对鸭卵巢组织玻璃化冷冻以及在鸭肾脏被膜下移植过程中的影响,试验分为5组(Con组:新鲜卵巢组织;0组、2.5组、5组、10组:0、2.5、5、10 mmol/L白藜芦醇),采用荧光定量PCR技术,检测玻璃化冷冻复苏后、复苏后鸭肾脏被膜下移植第3天和第9天卵巢组织中MST1、LATS1、YAP1和VEGF mRNA表达情况;HE染色观察移植后卵巢的卵泡形态。结果显示:与Con组相比,玻璃化冷冻显著降低卵巢组织MST1、LATS1、YAP1和VEGF mRNA相对表达量(P<0.05)。与0组相比,10 mmol/L白藜芦醇能显著增加玻璃化冷冻卵巢MST1、LATS1、YAP1和VEGF mRNA相对表达量(P<0.05)。卵巢移植3 d后,与0组相比,白藜芦醇能显著增加10组MST1、YAP1、VEGF,2.5组LATS1、YAP1,5组VEGF mRNA相对表达量(P<0.05);移植9 d后,白藜芦醇能显著降低MST1和LATS1 mRNA相对表达量(P<0.05)和显著升高YAP1和VEGF mRNA相对表达量(P<0.05)。白藜芦醇能显著增加冻融卵巢移植3 d和9 d的5组和10组正常卵泡率(P<0.05)。综上所述,白藜芦醇能提高鸭卵巢冻融移植后的卵泡存活率,以10 mmol/L为试验的最佳浓度。The purpose of this study was to investigate the effects of different concentrations of resveratrol on vitrification of duck ovarian tissue and subcapsular transplantation of duck kidney.The study contained 5 groups(Con group:fresh ovarian tis-sue;0,2.5,5,10 group:0,2.5,5,10 mmol/L resveratrol).Fluorescence quantitative PCR was used to detect the relative expres-sion of MST1,LATS1,YAP1 and VEGF mRNA in ovarian tissue after vitrification and resuscitation and subcapsular transplanta-tion of duckings kidney on the 3rd and 9th day.HE staining was used to observe the morphology of ovarian follicles after trans-plantation.The results showed that vitrification could significantly reduce the relative expression of MST1,LATS1,YAP1 and VEGF mRNA in ovary compared with Con group(P<0.05).Compared with 0 group,10 mmol/L resveratrol could significantly in-crease the relative expression of MST1,LATS1,YAP1 and VEGF mRNA in vitrified ovaries(P<0.05).Three days after ovarian transplantation,resveratrol significantly increased the relative expression of MST1,YAP1,VEGF in 10 group,LATS1,YAP1 in 2.5 group,and VEGF mRNA in 5 group compared with 0 group(P<0.05).Nine days after transplantation,resveratrol significantly decreased the relative expression of MST1 and LATS1 mRNA and increased the relative expression of YAP1 and VEGF mRNA(P<0.05).Resveratrol could significantly increase the normal follicular rate in groups 5 and 10 on the 3rd and 9th day of freeze-thawed ovarian transplantation(P<0.05).In conclusion,resveratrol improved follicle survival after freeze-thaw transplantation in duck ovaries,with 10 mmol/L as the optimal concentration for the test.
