摘  要:为研究广东省鸽源新城疫病毒(NDV)的遗传进化规律,试验于2021—2022年从广东6家疑似感染NDV的鸽场采集病鸽临床样品,进行RT-PCR诊断、病毒分离鉴定和动物回归试验,并对分离毒株进行生物学毒力测定以及F基因、HN基因的同源性和遗传进化分析。结果显示:共分离到6株鸽源NDV,对幼鸽进行分组攻毒14 d后,发病率均为100%,死亡率为60%~80%;6株毒株的F蛋白裂解位点氨基酸序列为112R-R-Q-K-R-F117/112R-R-R-K-R-F117,具有NDV强毒株的典型分子特征,和分离毒株ICPI的测定结果共同表明6株毒株均为强毒株;6株毒株均为NDV的ClassⅡ基因Ⅵ.2.1.1.2.2型;与传统疫苗株La Sota的同源性较低,且F、HN蛋白氨基酸出现多处位点变异。研究表明,广东省鸽源NDV流行以Class Ⅱ基因Ⅵ.2.1.1.2.2型为主,常用的疫苗株La Sota可能对广东地区流行的鸽源NDV保护效果不佳。In order to study the genetic evolution of Newcastle disease virus(NDV)from pigeon in Guangdong province,the clinical samples from diseased pigeons with suspected NDV infection in 6 pigeon farms in Guangdong province were diagnosed by RT-PCR,isolation and identification of virus and animal regression test from 2021 to 2022.Sequencing,biological virulence determination,homology and genetic evolution analysis of F gene and HN gene were carried out for the isolated strains.The re-sults showed that 6 strains of pigeon NDV were isolated after 14 days of challenge on grouped young pigeons,the morbidity was 100%,the mortality was 60%to 80%.In addition,the amino acid sequence of the cleavage site of F protein of 6 strains was 112R-R-Q-K-R-F117/112R-R-R-K-R-F117,which showed typical molecular characteristics of high virulent NDV strains.The re-sults of ICPI of 6 strains showed that all the 6 strains were strong strains.Six strains belonged to the classⅡgeneⅥ.2.1.1.2.2 type of NDV and had low homology with the traditional vaccine strain La Sota.The amino acids of F and HN proteins in 6 strains showed multiple site variations.It was suggested that the major genotype of pigeon NDV in Guangdong province was classⅡgeneⅥ.2.1.1.2.2,the commonly used vaccine strain La Sota might not be effective against the pigeon origin NDV currently prevalent in Guangdong province.
