摘 要:目的:探讨气管滴注苯并[b]荧蒽(BbFA)和二苯并[a,h]蒽(DahA)对成年雄性大鼠氧化应激和炎症因子的影响。方法:选取雄性SD大鼠48只,随机分成6组,分别为对照组(0 mg/kg),Bb FA低、高剂量组(分别为3×10^(-4)、9×10^(-4)mg/kg),Dah A低、高剂量组(分别为7×10^(-5)、2×10^(-4)mg/kg),Bb FA和Dah A联合组(3×10^(-4)mg/kg Bb FA+7×10^(-5)mg/kg Dah A),每组8只,每周气管滴注两次,连续染毒4周,观察不同剂量染毒组对大鼠肺灌洗液、肺组织与血清中氧化应激指标和炎症因子的影响。结果:与对照组相比,Dah A染毒组、Bb FA和Dah A联合组大鼠肺组织中SOD活力显著降低(P<0.05),且联合组中MDA、NO、i-NOS含量显著升高(P<0.05),其中MDA、i-NOS含量显著高于单独Bb FA或Dah A染毒组(P<0.05)。染毒组大鼠血清中氧化应激指标MDA含量均升高,且联合组MDA含量显著高于单独Bb FA或Dah A染毒的低剂量组(P<0.05)。与对照组比较,染毒组大鼠肺灌洗液和血清中炎症因子含量均升高,且联合组中IL-6含量显著高于单独Bb FA或Dah A染毒组(P<0.05)。结论:大鼠单独或是联合染毒Bb FA和Dah A后,可能是通过抑制SOD和CAT活性,升高MDA、NO和i-NOS活性引起体内氧化应激的改变,进而引起炎症因子IL-6水平的升高。OBJECTIVE:To investigate effects of benzo[b]fluoranthene(BbFA)and dibenzo[a,h]anthracene(DahA)exposure on oxidative stress and inflammatory factors in rats.METHODS:After 1 week of acclimatization,48 adult male SD rats were randomly divided into exposed and control groups.The rats were exposed to BbFA and DahA by intratracheal instillation twice a week in 4 weeks.Inflammatory factors in BALF and serum,and oxidative stress factors in lung tissue and serum were determined.RESULTS:Serum activity of SOD was significantly decreased in the DahA-exposed groups(P<0.05).Meanwhile,the BbFA+DahA exposed groups showed significantly increased levels of MDA,NO,i-NOS in the lung tissue compared with the control group(P<0.05),and the levels of MDA and i-NOS in the BbFA+DahA combined exposure group were significantly higher than that of the BbFA-exposed and the DahA-exposed groups(P<0.05).Additionaly,the content of MDA in the BbFA+DahA combined exposure group was significantly higher than low dose group of BbFA-exposed group and DahA-exposed group(P<0.05).The levels of IL-6 in the BbFA+DahA combined exposure group were significantly up-regulated compared with the BbFA-exposed and the DahA-exposed groups(P<0.05).CONCLUSION:Our results demonstrated that BbFA and DahA caused changes in inflammatory and oxidative factors that were related to inhibition of SOD activity and elevation of iNOS activity,and MDA,NO and IL-6 levels.
