摘 要:目的通过一测多评法同时测定甘草浸膏中甘草苷、芹糖异甘草苷、异甘草苷、甘草素和甘草酸铵5种有效成分的含量。方法采用高效液相色谱法对甘草浸膏中甘草苷、芹糖异甘草苷、异甘草苷、甘草素和甘草酸铵5种成分采用同一色谱条件进行分析:ChromCore AR-C_(18)色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.05%磷酸水溶液,梯度洗脱,流速为1.0 mL/min,柱温为25℃。以甘草酸铵为内标,计算其他4种成分的相对校正因子,并对不同仪器系统和色谱柱、不同体积流量、不同柱温、不同进样量进行了耐用性考察。最后用该一测多评法测定了甘草浸膏中5种有效成分的含量。结果在上述色谱条件下,5种成分在其线性范围内线性关系良好(r≥0.9999),平均加样回收率为92.39%~103.03%,RSD为1.45%~2.90%,RAD<2.0%。结论本研究所建立的一测多评法可同时测定甘草浸膏中甘草苷、芹糖异甘草苷、异甘草苷、甘草素和甘草酸铵5种有效成分的含量,该测定含量的方法准确可行,与外标法结果相当。Objective:The contents of five effective components,including glycyrrhizin,isoliquiritin apioside,isoliquiritin,liquiritigenin and ammonium glycyrrhizate,in liquorice extract were determined by one test and multiple assessment method.Methods:HPLC was used to analyze 5 components of iquorice extract,including glycyrrhizin,isoliquiritin apioside,isoliquiritin,liquiritigenin and ammonium glycyrrhizate under the same chromatographic condition:ChromCore AR-C_(18) column(4.6 mm×250 mm,5μm),mobile phase consisted of acetonitrile-0.05%phosphoric acid solution,gradient elution,flow rate was 1.0 mL/min.The column temperature was 25℃.Using ammonium glycyrrhizate as the internal standard,the relative correction factors of other 4 components were calculated,and the durability of different instrument systems and chromatographc columns,different volume flow,different column temperature and different sample size was investigated.Finally,the content of 5 effective components in liquorice extract was determined by the method of multiple assesmen.Results:Under the above mentioned chromatographic conditions,the linearity of the five components was good(r≥0.9999).The average recoveries were 92.39%-103.03%,RSD were 1.45%-2.90%,with RAD<2.0%.Conclusion:In this study,the multiple assessment method can simultaneously determine the content of 5 effective ingredients in liquorice extract,glycyrrhizin,isoliquiritin apioside,isoliquiritin,liquiritigenin and ammonium glycyrrhizate.The content determination method is accurate and feasible,and the results have no significant difference compared with the external standard method.
