摘 要:扩张蛋白通过诱导pH依赖性细胞壁延伸和应力松弛来促进细胞壁松驰。在高等植物的一系列生理发育过程中都需要扩张蛋白。本试验从羽衣甘蓝(Brassica oleracea L.var.acephala D.C.)基因组中克隆一个扩张蛋白基因BoEXPA2,对其生物信息学进行分析,并分别克隆到过表达和抑制表达载体中,以野生型作为对照,对转基因株系用GA、ABA及PAC处理进行发芽率及BoEXPA2表达量研究。结果表明,BoEXPA2蛋白与埃塞俄比亚芥、油菜、灰芥等的蛋白同源性最高。BoEXPA2基因表达量在发芽种子中显著提高,但干种子几乎没有表达。在发芽试验中,过表达株系发芽时间比野生型株系显著提前,而抑制表达株系的发芽时间与野生型株系相比显著延迟。外源GA增加了种子萌发过程中BoEPXA2的表达量,而外源ABA对BoEPXA2表达没有影响。用GA抑制剂PAC处理,抑制表达株系种子发芽率仍然显著低于野生型株系,而过表达株系的发芽受到PAC抑制的程度低于野生型株系。这些结果说明,BoEPXA2参与GA介导的种子萌发。Expansins are a cell wall protein that promotes cell wall loosening by inducing pH dependent cell wall elongation and stress relaxation.Expansins are required in a series of physiological development processes of higher plants.In this experiment,a gene of expansins,BoEXPA2 was cloned from the genome of Brassica oleracea,which was analyzed by bioinformaticsand and cloned into over-expression and inhibition expression vectors,respectively.The germination rate and BoEXPA2 expression of transgenic lines treated with GA,ABA and PAC were studied and wild type was used as control.The results showed that BoEXPA2 protein had the highest homology with Brassica carinata,Brassica napus and Hirschfeldia incana.The expression of BoEXPA2 gene significantly increased in germinated seeds,but almost not in dry seeds.In the germination test,the germination time of the overexpression line was significantly earlier than that of the wild type,while the germination time of the inhibition line was significantly delayed compared with that of the wild type.Exogenous GA increased the expression level of BoEPXA2 during seed germination,while exogenous ABA had no effect on the expression of BoEPXA2.When treated with GA inhibitor PAC,the seed germination rate of the inhibited expression line was still significantly lower than that of the wild type,while the germination of the over expression line was inhibited by PAC lower than that of the wild type.These results suggest that BoEPXA2 involved in seed germination mediated by GA.
