摘  要:为了明确8个彩色马铃薯新品系(P18、P22、P38、P126、P143、P218、P271和P359)在细胞遗传学和DNA分子水平上的遗传差异,本研究以彩色马铃薯紫彩3号为对照,采用染色体制片法,对各新品系的花粉育性、花粉母细胞PMCMⅠ染色体构型进行分析,并利用SSR标记技术构建了8个新品系的指纹图谱。结果表明,8个彩色马铃薯新品系的花粉育性依次为:55.32%,66.98%,47.32%,80.21%,60.12%,78.33%,49.34%和65.42%,其染色体配对构型依次为2n=4x=48=6.03Ⅰ+8.23Ⅱ+4.89Ⅲ+2.71Ⅳ、2n=4x=48=4.28Ⅰ+9.46Ⅱ+4.20Ⅲ+3.05Ⅳ、2n=4x=48=7.01Ⅰ+7.97Ⅱ+5.16Ⅲ+2.39Ⅳ、2n=4x=48=3.22Ⅰ+1.57Ⅱ+3.29Ⅲ+3.44Ⅳ、2n=4x=48=5.33Ⅰ+8.97Ⅱ+4.95Ⅲ+2.47Ⅳ、2n=4x=48=3.85Ⅰ+10.03Ⅱ+3.75Ⅲ+3.21Ⅳ、2n=4x=48=6.45Ⅰ+8.12Ⅱ+4.90Ⅲ+2.65Ⅳ和2n=4x=48=4.65Ⅰ+9.32Ⅱ+4.25Ⅲ+2.98Ⅳ,表明在细胞学水平上各品系间有一定差异。此外,利用筛选出的5对SSR适宜引物,对各新品系及对照材料的基因组DNA进行扩增,共获得74个SSR多态性标记,其多态性位点占比89.16%,进而以特异性引物STI046建立了8个彩色马铃薯新品系及对照紫彩3号的SSR指纹图,并以平均遗传距离(GD值)0.46为基准,将9个材料分为三类:新品系P22、P126、P143、P218、P271、P359、紫彩3号(ck)为一类,新品系P18和P38各单独为一类。In order to identify the genetic differences of eight new strains of colored potato P18,P22,P38,P126,P143,P218,P271 and P359 at the cytogenetic and DNA molecular levels,the chromosomal section method was used to analyze the pollen fertility and PMCMⅠchromosome configuration of pollen mother cells,and SSR marker technique were used to construct a SSR fingerprint map of eight new strains in the present research,with the control of potato material Zicai3.The results showed that the pollen fertility of the eight new strains of colored potato were 55.32%,66.98%,47.32%,80.21%,60.12%,78.33%,49.34%,and 65.42%,respectively.The chromosome pairing configuration of eight new strains were 2n=4x=48=6.03Ⅰ+8.23Ⅱ+4.89Ⅲ+2.71Ⅳ,2n=4x=48=4.28Ⅰ+9.46Ⅱ+4.20Ⅲ+3.05Ⅳ,2n=4x=48=7.01Ⅰ+7.97Ⅱ+5.16Ⅲ+2.39Ⅳ,2n=4x=48=3.22Ⅰ+1.57Ⅱ+3.29Ⅲ+3.44Ⅳ,2n=4x=48=5.33Ⅰ+8.97Ⅱ+4.95Ⅲ+2.47Ⅳ,2n=4x=48=3.85Ⅰ+10.03Ⅱ+3.75Ⅲ+3.21Ⅳ,2n=4x=48=6.45Ⅰ+8.12Ⅱ+4.90Ⅲ+2.65Ⅳ,and 2n=4x=48=4.65Ⅰ+9.32Ⅱ+4.25Ⅲ+2.98Ⅳ.The above results showed that there were some differences among the strains at the cytological level.In addition,five suitable SSR primers were screened to amplify genomic DNA of each new strain and the control material,a total of 74 SSR polymorphic markers were obtained,with the ratio of polymorphism loci of 89.16%.A SSR fingerprint map of eight new strains of colored potato and the control Zicai3 control was established based on the selected specific primer STI046.Using the average genetic distance(GD value)of 0.46 as the benchmark,the nine materials were divided into three categories:new strains P22,P126,P143,P218,P271,P359 as one and Zicai3(ck),P18,and P38 as one,respectively.
