摘 要:以人工加速老化的油菜种子为材料,利用傅里叶变换红外光谱(FT-IR)、二维相关红外光谱(2D-IR),结合曲线拟合、主成分分析(PCA)研究其营养物质变化,并对种子进行老化鉴别。结果表明,油菜种子中主要含有脂质、蛋白质、糖类等物质成分,不同人工老化程度油菜种子之间的FT-IR光谱吸收特性整体相似,吸收峰强比A_(1057)/A_(3382),A_(1656)/A_(3382),A_(1746)/A_(3382),A_(2926)/A_(3382)随老化时间的增加先减后增。3000~1000 cm^(-1)范围内(SD-IR)的吸收峰强度随老化时间的延长先减弱后增强。2D-IR显示,油菜种子随老化时间的增加在1800~1100 cm^(-1)范围内自动峰的数量、位置和强度发生明显变化;2990~2830 cm^(-1)范围内的曲线拟合分析结果显示,不同老化时间处理后的油菜种子的子峰(2854 cm^(-1),2925 cm^(-1))面积百分比随老化时间的增加发生变化。对1800~800 cm^(-1)范围内原始光谱进行PCA,结果显示,两种油菜种子前两个主成分分别获得98%和99%的累积方差贡献率,不同老化程度的样品均被100%正确分类。研究表明,FT-IR光谱结合统计分析方法,可以对人工老化油菜种子进行快速有效地检测和鉴别。Using artificially accelerated aging rapeseed seeds as materials,fourier transform infrared spectroscopy(FT-IR)and two-dimensional correlation infrared spectroscopy(2D-IR),combined with curve fitting and principal component analysis(PCA),were used to analyze the changes of nutrients,and the seeds were identified for aging.The results showed that the main contents of lipids,proteins,carbohydrate substances such as composition,different degree of artificial aging between rape seed FT-IR spectrum of light absorption properties in whole,absorption peak intensity than A_(1057)/A_(3382),A_(1656)/A_(3382),A_(1746)/A_(3382),A_(2926)/A_(3382)along with the increase of aging time after first decreased.In the range of 3000-1000 cm^(-1)(SD-IR),the absorption peak intensity decreased first and then increased with the aging time.The 2D-IR showed that the number,location and intensity of automatic peaks changed significantly with the increase of aging time from 1800 to 1100 cm^(-1).The results of curve fitting analysis in the range of 2990 to 2830 cm^(-1)showed that the percentage of subpeak area(2854 cm^(-1),2925 cm^(-1))of rapeseed seeds treated with different aging time changed with the increase of aging time.PCA was performed on the original spectrum in the range of 1800-800 cm^(-1),and the results showed that the first two principal components of the two kinds of rape seeds obtained 98%and 99%cumulative variance contribution,respectively,and the samples with different aging degrees were 100%correctly classified.The study showed that FT-IR spectra combined with statistical analysis method could be used to detect and identify artificially aged rape seeds quickly and effectively.
