摘　　要：目的 观察蒙药苏格木勒-3汤(SD-3)对大鼠酒精性心肌病(ACM)的保护作用,并探讨其机制。方法 制备酒精诱导的大鼠心肌病模型,共分为6组,分别为正常(CON)组、模型(ACM)组、SD-3低剂量(ACM+SL)组、SD-3中剂量(ACM+SM)组、SD-3高剂量(ACM+SH)组、阳性药物曲美他嗪(ACM+TI)组。计算心脏体质量比并观察SD-3对酒精诱导的心肌肥厚的影响,苏木素-伊红(HE)及Masson染色明确SD-3对ACM的保护作用。使用酶联免疫法测定肌酸激酶(CK-MB)及肌钙蛋白(CTn)T水平;生化仪测定血清三酰甘油(TG)、血清总胆固醇(TC)水平。Western印迹检测SD-3对酒精诱导的大鼠心肌损伤模型中B细胞淋巴瘤(Bcl)-2、Bcl-2相关X蛋白(Bax)、胱天蛋白酶(Caspase)3和Caspase7表达的影响。结果 与CON组相比,ACM组心脏体质量比、血清中CTnT、CK-MB、TG和TC含量及Bax、Caspase3、Caspase7表达显著升高,Bcl-2表达显著降低(P<0.05,P<0.01)。与ACM组相比,SD-3各剂量组心脏体质量比、血清中CTnT、CK-MB、TG和TC含量及Bax、Caspase3、Caspase7水平显著降低,Bcl-2水平显著升高(P<0.05,P<0.01)。结论 SD-3对ACM具有一定的保护作用,其机制可能与抑制细胞凋亡有关。Objective To observe the protective effect of mongolian medicine Sugemule-3 decoction(SD-3)on alcoholic cardiomyopathy(ACM)in rats and explore its mechanism.Methods The alcohol induced rat cardiomyopathy model was prepared,rats were divided into 6 groups,normal(CON)group,model(ACM)group,SD-3 low-dose(ACM+SL)group,SD-3 medium dose(ACM+SM)group,SD-3 high-dose(ACM+SH)group,and positive drug trimetazidine(ACM+TI)group,respectively.The heart to body mass ratio was calculated,and the effect of SD-3 on alcohol induced myocardial hypertrophy was observed.Hematoxylin-eosin(HE)and Masson staining were used to clarify the protective effect of SD-3 on ACM.Enzyme-linked immunosorbent assay was used to measure the levels of creatine kinase(CK-MB)and troponin(CTn)T.The biochemical analyzer was used to measure the levels of serum triglycerides(TG)and total cholesterol(TC).Western blot was used to detect the effects of SD-3 on the expressions of B-cell lymphoma(Bcl)-2,Bcl-2 related X protein(Bax),Caspase-3,and Caspase-7 in ACM rats.Results Compared with CON group,the heart to body mass ratio,serum levels of CTnT,CK-MB,TG,TC,and the expressions of Bax,Caspase3,Caspase7 were significantly increased,the expression of Bcl-2 was significantly decreased in ACM group(P<0.05,P<0.01).Compared with the ACM group,the heart to body mass ratio,serum levels of CTnT,CK-MB,TG,TC,and the levels of Bax,Caspase3,and Caspase7 were significantly decreased,the level of Bcl-2 was significantly increased in SD-3 dose groups(P<005,P<0.01).Conclusions SD-3 has a certain protective effect on ACM,and its mechanism might be related to inhibiting cell apoptosis.
