摘  要:目的评价藤茶总黄酮对大气细颗粒物(PM2.5)所致大鼠肺损伤的作用。方法雄性Wistar大鼠35只随机分为5组:对照组,模型组,藤茶总黄酮(TF)高、中、低剂量组,每组7只。模型组、TF各组气管滴注PM2.5混悬液10 mg/kg,每周1次,共4次,对照组则以生理盐水进行气管滴注。滴注次日起,对照组、模型组以0.5%羧甲基纤维素钠灌胃,TF组以不同剂量(500、250、125 mg/kg)藤茶总黄酮灌胃,每周5次,共4周。收集大鼠肺组织、肺泡灌洗液(BALF),观察肺组织病理形态,检测肺组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px),酶联免疫吸附试验(ELISA)检测BALF中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)。结果与对照组相比,模型组大鼠肺组织损伤明显,SOD、GSH-Px活性显著降低,MDA含量、BALF中TNF-α、IL-6、IL-1β水平显著升高。在中高剂量TF干预下,PM2.5肺损伤模型大鼠的肺组织损伤减轻,SOD、GSH-Px活性显著增强,MDA含量、BALF中TNF-α、IL-6、IL-1β水平显著下降,差异有统计学意义。结论藤茶总黄酮对PM2.5所致大鼠肺损伤具有一定的保护作用,这一作用与减轻大鼠肺部的氧化应激和炎症反应有关。Objective To evaluate the effect of tengcha flavonoids on lung injury of rats induced by atmospheric fine particulate matter(PM2.5).Methods 35 male Wistar rats were randomly divided into five groups:the control group,model group,tengcha flavonoids(TF)high-dose,medium-dose,and low-dose group,with 7 rats in each group.The model and TF groups were exposed to PM2.5 suspension by intratracheal instillation at a dose of 10 mg/kg once a week for four weeks,while the control group was instilled with normal saline.On the next day of modeling,the control and model groups were administered 0.5%sodium carboxymethyl cellulose solution,whereas the TF groups were given TF suspension in dosages of 500,250,and 125 mg/kg five times a week for four weeks.The lung tissues and bronchoalveolar lavage fluid(BALF)of rats were collected.The histopathological changes of lungs were observed.The levels of superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxidase(GSH-Px)in lung tissue were detected.The contents of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and interleukin-1β(IL-1β)in BALF were measured by enzyme-linked immunosorbent assay(ELISA).Results Compared to the control group,the model group showed obvious injury in lung tissues.The activities of SOD and GSH-Px in lung tissues were significantly decreased,the content of MDA in lung tissues and the levels of TNF-α,IL-6 and IL-1βin BALF were significantly increased in the model group.Under the treatment of TF with medium and high dose,the degree of lung injury was alleviated,the activities of SOD and GSH-Px were increased,the contents of MDA in lung tissues and the levels of TNF-α,IL-6 and IL-1βin BALF were decreased significantly.Conclusion Tengcha flavonoids has a certain protective effect on lung injury of rats induced by PM2.5,which is associated with reduction of oxidative stress and inflammatory response.
