非生物胁迫下BoEPXA2基因对羽衣甘蓝种子萌发的影响

摘  要:以野生型、BoEXPA2基因过表达和BoEXPA2基因抑制表达的羽衣甘蓝为试材,采用无机试剂NaCl、蔗糖(sucrose)和甘露醇(mannitol)分别设定低浓度和高浓度NaCl(100 mmol·L^(-1)和200 mmol·L^(-1))、蔗糖(150 mmol·L^(-1)和250 mmol·L^(-1))和甘露醇(200 mmol·L^(-1)或400 mmol·L^(-1)),分析在不同的胁迫下3个株系种子萌发情况,并测定各基因型不同胁迫下抗氧化酶活性,MDA含量及BoEXPA2的表达,以期探讨羽衣甘蓝BoEXPA2基因在非生物胁迫下对种子萌发的影响。结果表明:在非生物胁迫下,与野生型种子相比,BoEXPA2抑制表达种子的发芽率显著降低,而BoEXPA2过表达系的发芽率显著提高。在干旱胁迫和盐胁迫下,3种基因型种子的超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量均有增加,但是BoEXPA2过表达株系种子的SOD、POD、CAT 3种酶活性远高于对照和抑制表达株系,而MDA含量低于对照和抑制表达株系;当种子被NaCl、蔗糖和甘露醇处理时,随着含量的增加,BoEXPA2的表达程度均会随之降低,这表明非生物胁迫抑制了发芽种子中BoEXPA2的表达。The wild type,BoEXPA2 gene overexpression,and BoEXPA2 gene suppressed expression of kale were used as experimental materials.Three inorganic reagents,NaCl,sucrose,and mannitol were used.Low and high concentrations of NaCl(100 mmol·L^(-1) and 200 mmol·L^(-1)),sucrose(150 mmol·L^(-1) and 250 mmol·L^(-1)),and mannitol(200 mmol·L^(-1)and 400 mmol·L^(-1))groups were set,respectively.The seed germination of three strains under different stresses was analyzed,and antioxidant enzyme activity,MDA content,and BoEXPA2 expression were measured for each genotype under different stresses,in order to explore the effect of BoEXPA2 gene on seed germination of kale under abiotic stress.The results showed that under abiotic stress,compared with wild type seeds,the germination rate of BoEXPA2 inhibited expression seeds was significantly reduced,while the germination rate of BoEXPA2 overexpression lines was significantly increased.Under drought stress and salt stress,the activities of superoxide dismutase(SOD),peroxidase(POD),catalase(CAT)and the content of malondialdehyde(MDA)in the seeds of the three genotypes were increased,but the activities of SOD,POD,CAT in the seeds of the BoEXPA2 overexpression strain were far higher than those of the control and suppression expression strains,while the activities of MDA were lower than those of the control and suppression expression strains.When the seeds were treated with NaCl,sucrose and mannitol,the expression of BoEXPA2 decreased with the increase of their content.The results indicated that abiotic stress inhibits the expression of BoEXPA2 in germinated seeds.
