摘 要:为明确国内主产区不同主栽石榴品种的遗传多样性,构建DNA指纹图谱,本研究以30个石榴品种为试材,利用琼脂糖凝胶电泳和毛细管电泳筛选多态性引物,建立了石榴品种SSR分子标记鉴定体系,然后利用样品信息与指纹代码配置组合的串联代码,构建了不同石榴品种的DNA指纹图谱。结果表明,从36对引物中筛选出扩增产物条带清晰稳定、多态性高的引物10对,共检测出48个等位基因,平均为4.8个,观察杂合度和期望杂合度平均分别为0.50和0.60,Shannon’s指数和Nei’s多样性指数平均分别为1.11和0.59,多态信息含量在0.33~0.72之间,平均为0.53。聚类分析发现,30个品种被分为3组,A组包括16个品种,B组包括8个品种,C组包括6个品种。基于样品、引物和等位基因信息,构建了30个品种的特异指纹代码及指纹图谱二维码。本研究结果可为石榴种质资源分子鉴定及开发利用提供理论依据。In order to identify the genetic diversity of major pomegranate varieties and construct their DNA fingerprints,30 pomegranate varieties were used as materials,the agarose and capillary electrophoresis were used to screen polymorphic primers,and the SSR molecular marker identification system were construc-ted.Then the DNA fingerprints of the pomegranate varieties were constructed by using the tandem codes con-taining sample,primer and allele information.The results showed that ten pairs of primers with clear and sta-ble bands and high polymorphism were selected from 36 pairs of primers.Using them,48 alleles were detected with the mean of 4.8 per every pair of primer.The average observed heterozygosity and expected heterozygosity were 0.50 and 0.60,respectively.The Shannon’s index and Nei’s diversity index were 1.11 and 0.59,re-spectively.The polymorphism information content ranged from 0.33 to 0.72,with the mean of 0.53.The cluster analysis results showed that the 30 varieties could be classified into 3 groups,among which,the group A in-cluded 16 varieties,the group B included 8 varieties,and the group C included 6 varieties.Based on the in-formation of samples,primers and alleles,the specific fingerprint codes and their two-dimensional codes of the 30 varieties were constructed.The results of this study could provide theoretical bases for molecular identifica-tion,development and utilization of germplasm resources of pomegranate.
