摘 要:为明确不同寄主上的烟粉虱体内共生菌多样性的差异,本研究构建了取食番茄和棉花的两个烟粉虱种群,利用PacBio测序平台对其体内共生菌的多样性及优势共生菌的感染情况进行分析。结果表明,取食番茄的烟粉虱体内优势共生菌为Portiera、Rickettsia、Hamiltonella;寄主转为棉花后,共生菌的种类和扩增子序列变体(amplicon sequence variants, ASVs)数量明显减少,物种丰富度降低,优势菌群增加为Portiera、Rickettsia、Hamiltonella、Cardinium 4种,且相对丰度发生了显著变化。取食两种寄主的烟粉虱体内感染率最高的共生菌均为Protiera,其次为Rickettsia、Hamiltonella和Cardinium,并且以3~4种共生菌协同感染为主;寄主由番茄转为棉花后,共生菌协同感染的丰富度明显增加。由此可见,寄主转移对烟粉虱体内共生菌的多样性及感染形式均有显著影响,该结果可为研究烟粉虱-共生菌-寄主三者之间的协同进化关系提供理论依据。In order to make clear the differences in endosymbiotic bacteria diversity of Bemisia tabaci in different host plants,two B.tabaci populations fed with tomato and cotton were established in this experiment.The diversity of endosymbiotic bacteria in the adults of the two B.tabaci populations and the infection condi-tion of dominant symbiotic bacteria were determined by PacBio sequencing platform.The results showed that the dominant symbiotic bacteria in B.tabaci population fed with tomato were Portiera,Rickettsia and Hamil-tonella.The species of symbiotic bacteria and the number of amplicon sequence variants(ASVs)decreased obviously when B.tabaci were transferred to cotton from tomato,and the dominant symbiotic bacteria increased to four genus as Portiera,Rickettsia,Hamiltonella and Cardinium with the relative abundance changing signifi-cantly.The highest infection rate of Protiera was the highest in the two B.tabaci populations,followed by Rick-ettsia,Hamiltonella and Cardinium,andthe co-infectionof three or four species of symbiotic bacteria was the main form.The co-infection richness of symbiotic bacteria increased significantly when B.tabaci were trans-ferred to cotton from tomato.In conclusion,host conversion had significant effect on the diversity of endo sym-biotic bacteria in B.tabaci.These results could provide theoretical bases for researching the co-evolutionary re-lationship between B.tabaci,endosymbiotic bacteria and host plants.
