摘  要:为探讨马立克病毒(Marek’s disease virus, MDV)与肿瘤发生、发展的关系,本研究建立了鸡马立克病(Marek’s disease, MD)模型,并利用病理组织学、免疫组织化学、荧光定量PCR、酶联免疫吸附(ELISA)等方法测定了肿瘤相关蛋白热休克蛋白60(heat shock protein 60, HSP60)在鸡肺脏组织内的定位及转录水平和含量的动态变化。结果显示,HSP60在攻毒组鸡肺脏肿瘤细胞的细胞质内强表达,试验周期内攻毒组HSP60 mRNA转录水平极显著高于对照组(P<0.01),总体呈开口向下的抛物线型,即7~28日龄间持续升高,28日龄达到最高值,分别为空白对照组和疫苗对照组的1.39倍和1.44倍,之后降低;HSP60含量7~28日龄攻毒组极显著高于空白对照组(P<0.01),35日龄后两者间差异不显著。该结果为进一步研究HSP60在肿瘤发生过程中的生物学作用奠定了基础。In order to investigate the relationship between Marek’s disease virus(MDV)and tumorigene-sis and development,a chicken Marek’s disease(MD)model was established in this study,and the localiza-tion,transcription and expression levels of the tumor-associated protein heat shock protein 60(HSP60)in chicken lung tissues were determined by using histo pathology,immunohistochemistry,fluorescence quantita-tive PCR and enzyme-linked immunosorbent assay(ELISA)methods.The results showed that HSP60 was strongly expressed in the cytoplasm of chicken lung tumor cells in the takedown group.The HSP60 mRNA transcript level in the takedown group was extremely significantly higher than that in the control group during the test cycle(P<0.01),and showed an overall parabolic pattern with a downward opening,namely,contin-ued to increase during 7 to 28 days of age,and reached the maximum at the 28 days of age,which was 1.39 and 1.44 folds of that of the blank and vaccine control groups,respectively,after that,it decreased.The ex-pression of HSP60 was extremely significantly higher in the takedown group than that in the blank control group from 7 to 28 days of age(P<0.01),and the difference between them was not significant after 35 days of age.These results could lay a foundation for further research on the biological role of HSP60 in tumorigenesis.
