子痫前期胎盘中MiR-26b的表达量及其与血管新生、细胞侵袭的相关性

摘  要:目的:探讨子痫前期胎盘中MiR-26b的表达量及其与血管新生、细胞侵袭的相关性。方法:选取2022年1—12月于中山火炬开发区人民医院分娩的130例子痫前期产妇作为子痫前期组,根据病情严重程度将产妇分为轻度子痫前期组(n=97)、重度子痫前期组(n=33);选取同期于本院分娩的健康产妇50例作为正常对照组。对比三组胎盘组织中MiR-26b表达量,血管新生、细胞侵袭相关表达量的差异,分析子痫前期产妇胎盘组织中MiR-26b表达量与血管新生、细胞侵袭的相关性。结果:轻度子痫前期组、重度子痫前期组胎盘组织中MiR-26b表达量高于正常对照组,胎盘组织中白血病抑制因子(LIF)、血管内皮生长因子(VEGF)、基质金属蛋白酶-9(MMP-9)、胎盘生长因子(PLGF)、醋酸激酶1(ACK1)、同源盒蛋白B3(HOXB3)、染色质SWI/SNF相关基质关联肌动蛋白依赖性调节因子亚家族A5(SMARCA5)、层粘连蛋白α4(LAMA4) mRNA表达量低于正常对照组,其中重度子痫前期组上述指标表达量变化幅度更大,差异有统计学意义(P<0.001)。子痫前期胎盘组织中MiR-26b表达量与血管新生、细胞侵袭相关基因表达量均呈负相关(P<0.05)。结论:子痫前期胎盘组织中MiR-26b表达量异常增高,可能通过抑制血管新生、细胞侵袭相关基因表达而参与疾病病情进展。Objective:To investigate the expression of MiR-26b in preeclampsia placenta and its correlation with angiogenesis,cell invasion.Method:A total of 130 pregnant women with preeclampsia who gave birth in Zhongshan Torch Development Zone People's Hospital from January to December 2022 were selected as preeclampsia group,the pregnant women were divided into the mild preeclampsia group(n=97)and the severe preeclampsia group(n=33)according to the severity of the disease;fifty healthy pregnant women who gave birth in our hospital during the same period were selected as the normal control group.The differences of MiR-26b expression,angiogenesis and cell invasion related expression in placental tissues of three groups were compared,and the correlation between MiR-26b expression in placental tissues of preeclampsia women and angiogenesis and cell invasion were analyzed.Result:The expressions of miR-26b in placental tissue of the mild preeclampsia group and the severe preeclampsia group were higher than that of the normal control group,the mRNA expression levels of leukemia inhibitory factor(LIF),vascular endothelial growth factor(VEGF),matrix metallo-proteinase 9(MMP-9),placental growth factor(PLGF),acetate kinase 1(ACK1),homeobox protein B3(HOXB3),chromatin SWI/SNF-related matrix-associated actin-dependent regulatory factor subfamily A5(SMARCA5)and lamininα4(LAMA4)were lower than those in the normal control group,and the expression levels of the above indicators in placental tissue of the severe preeclampsia group had a greater variation,the differences were statistically significant(P<0.001).The expression of MiR-26b in placenta tissue of preeclampsia were negatively correlated with the expression of genes related to angiogenesis and cell invasion(P<0.05).Conclusion:The abnormally high expression of MiR-26b in placenta of preeclampsia may participate in the progression of the disease by inhibiting the expression of genes related to angiogenesis and cell invasion.
