摘 要:目的 研究并建立贯叶金丝桃UPLC指纹图谱和多指标成分的含量测定方法。方法 采用Eclipse Plus C18色谱柱(100 mm×2.1 mm,1.8μm);以乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱,检测波长为300 nm;流速为0.25 mL·min^(-1);柱温为25℃。建立了同时测定金丝桃苷、芦丁、槲皮苷、槲皮素含量及指纹图谱的方法,并结合相似度评价、聚类分析、主成分分析和正交偏最小二乘法-判别分析(OPLS-DA)对22批贯叶金丝桃样品进行质量分析。结果 建立了贯叶金丝桃药材UPLC指纹图谱共有模式,标定了11个共有峰,并指认了其中9个,主成分分析基本可将22批贯叶金丝桃按照不同产地分为5类;采用OPLS-DA分析,得到5个质量差异标志物,差异排序依次为峰10(槲皮苷)>峰5>峰6(芦丁)>峰8(异槲皮苷)>峰1(绿原酸)。含量测定结果表明,野生品与栽培品4个指标成分总含量差异不甚明显,不同产地4个指标成分总量略有差异,其中以陕西、新疆产地的含量较高。结论 建立的UPLC指纹图谱和4个指标成分同时测定的方法可用于贯叶金丝桃药材的质量控制及质量标准提升。Objective To establish a UPLC fingerprints and multi-component quantitation of hyperici perforati herba.Methods Eclipse Plus C18(100 mm×2.1 mm,1.8μm)column was used.The mobile phase was acetonitrile-0.1%phosphoric acid solution with gradient elution.The detection wavelength was 300 nm,the flow rate was 0.25 mL·min^(-1) and the column temperature was 25℃.The content of hyperoside,rutin,quercitrin,quercetin was determined and the fingerprint was established.Variance analysis of single factor,combined with cluster analysis(CA),principal component analysis(PCA)and the partial orthogonal least square discriminant analysis(OPLS-DA)was used to analyze the quality 22 batches of hyperici perforati herba.Results The common pattern of UPLC fingerprints was established.We distinguished out 11 common peaks.CA and PCA showed that 22 batches of different origins of hyperici perforati herba were broadly divided into 5 categories.The OPLS-DA showed 5 difference markers,following the order of significance difference:peak 10(quercitrin)>peak 5>peak 6(rutin)>peak 8(isoquercitrin)>peak 1(chlorogenic acid).There was no big difference in the wild and cultivated products as to the content of 4 ingredients.The content of the 4 indicators were slightly different from different origins,the content of Shaanxi and Xinjiang production areas was higher.Conclusion The fingerprint and content determination method can be used to control the quality and improve the quality standard of the hyperici perforati herba.
