摘 要:目的研究银翘散对铜绿假单胞菌群感效应分子和生物膜的影响。方法通过倍比稀释法检测银翘散对铜绿假单胞菌的最低抑菌浓度;采用CV026平板显色反应观测铜绿假单胞菌分泌群感效应信号分子;采用革兰染色法观察银翘散对生物膜形态的影响;采用实时荧光定量PCR法分析银翘散对铜绿假单胞菌群感效应基因和生物膜分散基因的表达的影响。结果银翘散对铜绿假单胞菌的最小抑菌浓度是500 mg/ml;银翘散对群感效应信号分子的分泌有抑制作用;银翘散可抑制铜绿假单胞菌生物膜的形成;银翘散可下调对群感效应系统基因和铜绿假单胞菌膜分散基因的表达水平。结论银翘散通过抑制铜绿假单胞菌群感效应系统,达到抗铜绿假单胞菌生物膜作用。Objective To investigate the effects of Yinqiao powder on the quorum sensing molecules and biofilms of Pseudomonas aeruginosa.Methods The study used the minimum inhibitory concentration(MIC)method to detect the minimum bacteriostatic concentration of Yinqiao powder on Pseudomonas aeruginosa.The purple rod bacterial plate color reaction was used to observe the secretion of quorum sensing signal molecules by Pseudomonas aeruginosa.Gram staining was used to observe the effects of the drug on the morphology of biofilms.QRT-PCR was used to analyze the changes in gene expression of quorum sensing system and biofilm dispersal genes in Pseudomonas aeruginosa.Results The minimum bacteriostatic concentration of Yinqiao powder on Pseudomonas aeruginosa was 500 mg/ml,and Yinqiao powder had an inhibitory effect on quorum sensing signal molecules.Yinqiao powder could inhibit the formation of Pseudomonas aeruginosa biofilm;Yinqiao powder downregulated the expression levels of the quorum sensing system genes and Pseudomonas aeruginosa biofilm dispersal genes.Conclusion Yinqiao powder exerts an anti-biofilm effect on Pseudomonas aeruginosa by inhibiting the quorum sensing system.
