摘 要:为探究进口高油大豆在苏式仓储藏过程中真菌群落的演替变化,选取当年收获的高油大豆进入苏式仓中过夏储藏,储藏周期3个月,定期取样进行水分测定,利用Illumina Miseq高通量测序技术分析原始状态和其他储藏阶段大豆中真菌群落的演替。结果表明:不同储藏时期的大豆样品中的真菌群落共计属于4个门,9个纲,14个目,15个科,21个属,19个种。大豆在不同储藏时期的4个优势菌门为子囊菌门(Ascomycota)、担子菌门(Basidiomycota)、羊肚菌门(Mortierellomyco)、毛霉菌门(Mucoromycota)。在种的水平上进行分类统计分析发现,相对丰富含量较高的10个菌种在数量上均呈现随着储藏时间的延长而有不同程度的增加趋势(C47>B47>A47),并发现了1种检疫性病原真菌大麦叶烧病菌(Mycosphaerella tassiana)和1种有害真菌赤霉菌(Gibberlla)。通过Ven图分析和Beta多样性分析发现,前两个储藏期的大豆样品中真菌群落结构具有较高的相似度,而到了第三储藏期与前两者真菌群落结构差异较大,说明到了第三储藏期大豆中的真菌群落不仅多样性显著增加,真菌群落结构也发生了明显演替。In order to dynamicing monitor the state of fungal community of imported high oil soybean during storage,Illumina MiSeq high-throughput sequencing was used to analyze the successive changes of fungal community of the imported high oil soybean harvested this summer which placed in Soviet-style warehouse for over-duning storage in 3 months.The result showed that the fungal communities of the high oil soybean in different storage periods belonged to 4 phyla,9 classes,14 orders,15 families,21 genera and 19 species.The four dominant phyla of soybean in different storage periods were Ascomycota,Basidiomycota,Mortierellomyco and Mucoromycota.At the species level,classification statistical analysis showed that there were 10 strains with relatively high abundance presenting an increasing trend with the extension of storage time(C47>B47>A47),and a quarantine pathogenic fungus Mycosphaerella tassiana and a harmful fungus Gibberella were found.Through the analysis of Ven map and Beta diversity,it was found that the fungal community structure of soybean samples in the first two storage periods were relatively similar,but in the third storage period was significantly different,it indicated that the fungal community diversity of soybean samples in the third storage period not only increased significantly,but also experienced obvious succession of fungal community structure.
