摘 要:目的探讨鸢尾素对肾细胞癌的影响及PGC-1α、UCP-1在其中发挥的关键性作用。方法采用Western blot测定鸢尾素对人肾细胞腺癌细胞(ACHN)PGC-1α、UCP-1蛋白的影响;采用实时荧光定量逆转录PCR(qRT-PCR)方法检测鸢尾素对其自噬相关和褐变相关mRNA的影响;采用油红O染色检测鸢尾素对噬脂的影响;采用凋亡试剂盒检测鸢尾素对细胞凋亡率的影响;采用划痕实验检测鸢尾素对细胞的愈合和修复能力。结果鸢尾素能够上调棕色化相关蛋白(PGC-1α、UCP-1)及mRNA(PGC-1α、UCP-1、PRDM16)的表达并激活自噬相关mRNA(Beclin-1、LC3Ⅱ、P62)的表达(P<0.05);鸢尾素能够促进噬脂,抑制ACHN细胞增殖、细胞迁移运动与修复能力(P<0.05)。结论鸢尾素能够通过激活PGC-1α、UCP-1促进噬脂和褐变进而抑制ACHN细胞增殖和迁移。Objective To investigate the effects of irisin on renal cell adenocarcinoma and the key roles of PGC-1αand UCP-1 on it.Methods The effects of irisin on PGC-1αand UCP-1 proteins of human renal cell adenocarcinoma cell(ACHN)were determined by Western blot.The effects of irisin on autophagy and browning related mRNA were detected by real-time fluorescent quantitative reverse transcription PCR(qRT-PCR).The effect of irisin on lipophagy was detected by oil red O staining.The effect of irisin on apoptosis rate was detected by apoptosis kit.The healing and repair ability of irisin on cells was tested by scratch test.Results Irisin could up-regulate the expression of browning-related protein(PGC-1α,UCP-1)and mRNA(PGC-1α,UCP-1,PRDM16)as well as activate the autophagy-related mRNA(Beclin-1,LC3Ⅱ,P62,P<0.05).Irisin could promote lipophagy,inhibit the proliferation of ACHN cells,and the migration and repair ability of tumor cells(P<0.05).Conclusion Irisin can inhibit the proliferation and migration of renal cell ACHN cells by activating PGC-1αand UCP-1 to promote lipophagy and browning.
