人工老化和自然老化家榆种子的iTRAQ蛋白质组学分析

摘  要:种子老化是指种子成熟后活力逐渐下降的过程。自然老化是衡量家榆种子耐储藏性的直接指标,但因其过程缓慢,所以在科学研究中常用人工加速老化代替。为了探究人工老化与自然老化在分子机理上的异同,本研究采用iTRAQ(Isobaric tags for relative and absolute quantitation)法分别测定了家榆种子人工老化和自然老化早期的蛋白质,并对两种老化方式中的差异蛋白质数量进行比较。结果显示,人工老化后丰度差异蛋白有79个,自然老化后丰度差异蛋白有102个,这些差异蛋白在GO功能注释和KEGG通路注释上没有显著不同。其中,39个蛋白在人工老化和自然老化中均为差异蛋白且趋势一致,1个蛋白在两种老化方式下丰度变化趋势相反。对两种老化方式中不同的差异蛋白分析发现,39个蛋白仅在人工老化后丰度发生变化,61个蛋白仅在自然老化后丰度变化。对这些不同的差异蛋白进行分析,发现他们的GO功能注释和KEGG通路注释没有显著差异,表明人工加速老化和自然老化具有相似的分子变化谱。Seed ageing is the gradual decline of seed vigor.Natural aging is a direct indicator of the storage resistance in seeds,but because of its slow process,artificial accelerated aging is commonly used in scientific research instead.In order to explore the similarities and differences in molecular mechanism between artificial aging and natural aging.In this study,iTRAQ method was used to determine the proteomics of Ulmus pumila seeds in the early stage of artificial aging and natural aging,and then the ageing-related proteins between these two groups were compared.The results showed that there were 79 proteins in artificial aging and 101 proteins in natural aging with variant abundance.These proteins are not significantly different in GO function annotation and KEGG pathway annotation.Among them,the abundance of 39 proteins both changed in artificial aging and natural aging with the same tendency,and the abundance of one protein increased in artificial aging but decreased in natural aging.Then we analyzed the different proteins with variant abundance between the two aging methods.The results showed that 39 proteins with variant abundance were only found in the artificial aging,and 61 proteins were only found in the natural aging.However,it is found that their GO function annotations and KEGG pathway annotations are of no significant difference,suggesting a similar change in molecular profile between artificial and natural aging.
