摘 要:中医药具有悠久的历史,中医在防治疾病的临床过程中积攒下来丰富的经验,形成了中国独有的中医药理论体系。中医药是中华民族的瑰宝,在治疗复杂性疾病方面具有独特的优势,但是中药由于其组成复杂,靶点多样,如何快速评价药效、精准高效地发现中药活性成分、优化组分配伍并明确作用靶点是中药现代化研究中的难点。高内涵筛选技术作为一项高通量、多靶点、多通道以及全自动化采集荧光图像并进行分析的技术,能够高通量地获取并分析细胞对感兴趣药物所产生的多维生物效应,非常适用于中药现代化研究中多层次多靶点地评价药效、筛选有效成分、诠释配伍作用、优化组分配伍、探究药理机制、建立中药潜在损伤预警体系保障用药安全以及评价中药质量。本文简要介绍了高内涵筛选技术及其目前的发展状况,总结了高内涵筛选技术在中药现代化研究中的应用进展,展望了高内涵筛选结合3D细胞培养、微流控芯片、神经网络等新兴技术助力中药现代化研究的新方向。Traditional Chinese medicine(TCM)boasts a long history and a distinctive theoretical system developed through the accumulated clinical experience in disease prevention and treatment.While TCM offers unique advantages in treating complex diseases,its complex composition and diverse targets pose challenges in rapidly evaluating efficacy,precisely and efficiently discovering active ingredients,optimizing component combinations,and elucidating target actions in the modernization research of TCM.High-content screening(HCS)technology,with its high-throughput,multi-target,multi-channel,and fully automated capabilities for collecting and analyzing fluorescent images,can rapidly obtain and analyze the multidimensional biological effects of cells in response to drugs of interest.It is highly suitable for evaluating the efficacy of TCM at multiple levels and targets,screening effective components,interpreting compatibility effects,optimizing component combinations,exploring pharmacological mechanisms,establishing a toxicological early warning system for TCM safety,and evaluating the quality of TCM.This article briefly introduces the concept and current development status of HCS technology,summarizes its applications and progress in the modernization research of TCM,and envisions new directions by combining HCS with emerging technologies such as 3D cell culture,microfluidic chips,and neural networks to facilitate TCM modernization researc.
