摘 要:目的研究骆驼刺Alhagi sparsifolia地上部分的化学成分。方法采用多种色谱技术进行分离纯化,根据其理化性质和波谱数据对化合物结构进行鉴定,并通过MTT法测试各化合物抗人宫颈癌He La细胞的活性。结果从骆驼刺乙醇提取物二氯甲烷部位中分离得到12个化合物,分别鉴定为(2S,3S)-methylhexahydro-3H-pyrrolo[NH,2-a]imidazol-1-one(1)、5,6-二羟基-7-甲氧基黄酮(2)、汉黄芩素(3)、2-亚乙基-3-甲基琥珀酸(4)、3,4′-二羟基-3′-甲氧基苯丙酮(5)、calendin(6)、N-苯乙基乙酰胺(7)、对甲氧基苯甲酸(8)、黄芩新素Ⅱ(9)、4-羟基苯乙醇(10)、3′-甲氧基大豆苷元(11)、松脂素(12)。结论化合物1为新化合物,命名为骆驼刺碱;且化合物2~11为首次从该植物中分离得到,化合物1、3、9、11具有宫颈癌HeLa细胞毒活性,半数抑制浓度(median inhibition concentration,IC50)分别为20.20、35.25、49.11、48.73μmol/L。Objective To study the chemical composition of the above-ground parts of Alhagi sparsifolia.Methods Various chromatographic techniques were used for the separation and purification,the structures of the compounds were identified based on their physicochemical properties and spectral data,and the compounds were tested for their activity against human cervical cancer cells by MTT assay.Results Twelve compounds were isolated from the dichloromethane site of the ethanol extract of A.sparsifolia and identified as(2S,3S)-methylhexahydro-3H-pyrrolo[NH,2-a]imidazol-1-one(1),neglectein(2),wogonin(3),2-ethylidene-3-methylbutanedioic acid(4),3,4′-dihydroxy-3′-methoxyphenylacetone(5),calendin(6),N-phenylethyl acetamide(7),p-methoxybenzoic acid(8),skullcaflavone II(9),4-hydroxyphenethyl alcohol(10),3′-methoxydaidzein(11),pinoresinol(12).Conclusion Compound 1 is a new compound named alhagine,and compounds 2—11 were isolated from the plant for the first time,compounds 1,3,9 and 11possessed cervical cancer HeLa cytotoxic activity with IC50 of 20.20,35.25,49.11,48.73 μmol/L,respectively.
