摘 要:目的:研究天马颗粒对裸鼠结肠癌移植瘤组织中RAGE、CD34、D2-40表达的影响探讨天马颗粒抑癌机制。方法:以SPF级雌雄各半裸鼠为研究对象,通过皮下种植HCT116细胞株构建裸鼠移植瘤模型,随机将其分为三组:空白对照组,天马颗粒低浓度组(5.39g/kg)、天马颗正常浓度组(10.79g/kg),种植瘤形成后施行药物干预,然后摘取结肠癌组织,进行免疫组化染色检测,观察RAGE、CD34、D2-40蛋白表达量。结果:同空白对照组相比,天马颗粒低浓度组、天马颗粒正常浓度组大肠癌组织中RAGE蛋白具有明显差异(P<0.05),CD34天马颗粒组较空白对照组有明显差异(P<0.05),D2-40天马颗粒组较空白对照组有明显差异(P<0.05)。结论:天马颗粒可以抑制RAGE蛋白、CD34、D2-40蛋白表达,改善肿瘤炎症微环境,抑制肿瘤血管、淋巴管生成。Objective:To study the effect of Tianma Granules on the expression of RAGE,CD34 and D2-40 in colon cancer transplanted tumor tissues of nude mice and to explore the tumor suppressive mechanism of Tianma Granules.Methods:SPF-grade male and female half-nude mice were used as the research subjects.A nude mouse transplanted tumor model was constructed by subcutaneously planting HCT116 cell lines.They were randomly divided into three groups:blank control group,Tianma granule low concentration group(5.39 g/kg),In the Tianma Granule normal concentration group(10.79 g/kg),drug intervention was performed after the formation of implanted tumors,and then the colon cancer tissue was removed and immunohistochemical staining was performed to observe the expression levels of RAGE,CD34,and D2-40 proteins.Results:Compared with the blank control group,there were significant differences in RAGE protein in colorectal cancer tissues between the Tianma Granule low-concentration group and the Tianma Granule normal-concentration group(P<0.05).The CD34 Tianma Granule group was significantly different from the blank control group(P<0.05).The D2-40 Tianma granule group was significantly different from the blank control group(P<0.05).Conclusion:Tianma granules can inhibit the expression of RAGE protein,CD34,and D2-40 proteins,improve the tumor inflammatory microenvironment,and inhibit the formation of tumor blood vessels and lymphangiogenesis.
