摘 要:目的:基于“平衡理论”从调节骨质疏松大鼠模型骨代谢与骨转化平衡探讨补肾益骨方抗骨质疏松的作用机制。方法:选取8月龄SPF级SD大鼠60只随机分为空白组,模型组,对照组,高、中、低剂量组(n=10)。所有大鼠均通过去双侧卵巢手术造模,造模2周对照组予阿伦磷酸钠灌胃,模型组予生理盐水等量灌胃,高、中、低剂量组分别根据分组使用不同剂量补肾益骨方药液灌胃。药物干预持续12周,取大鼠股骨中段骨质HE检测股骨病理改变,采集大鼠静脉血液检测血清中血Ca、P浓度,ELISA法检测PINP、PICP、CTX、NTX表达水平。结果:H染色检查显示模型组骨组织呈明显骨质疏松改变。药物干预后大鼠骨质疏松病理改变明显改善;模型组大鼠血磷、血钙浓度低于空白组(P<0.05),药物干预后血磷、血钙浓度有所上升(P>0.05);模型组大鼠PINP、PICP、CTX、NTX浓度较空白组均明显升高(P<0.05),对照组及低、中、高剂量组大鼠PINP、PICP、CTX、NTX浓度较模型组降低(P<0.05);低、中、高剂量组大鼠PINP、PICP、CTX、NTX浓度较对照组有不同程度的降低(P<0.05),中剂量组大鼠PICP、CTX、NTX浓度与空白组比较无统计学差异(P>0.05)。结论:补肾益骨方能改善去卵巢骨质疏松大鼠模型骨质,机制与调节鼠钙、磷代谢,遏制骨代谢亢盛,降低骨转化率有关,不同剂量“补肾益骨方”无用药剂量依赖趋势,以中等剂量调节骨代谢效果明显。Objective:To explore the anti-osteoporosis mechanism of Bushen Yigu prescription by regulating bone metabolism and bone transformation balance in osteoporosis rat model based on“balance theory”.Methods:608-month-old SPF SD rats were randomly divided into blank group,model group,control group,high,medium and low dose groups(n=10).All rats underwent bilateral ovarian removal surgery to create a model.The control group was intragastrically administered with sodium alendronate for 2 weeks,the model group was intragastrically administered with an equal amount of normal saline,and the high,medium,and low-dose groups were administered different doses according to the group.The kidney-tonifying and bone-reinforcing prescription liquid is administered into the stomach.The drug intervention lasted for 12 weeks.HE was taken from the middle part of the femoral bone of rats to detect the pathological changes of the femur.The venous blood of the rats was collected to detect the blood Ca and P concentrations in the serum.The expression levels of PINP,PICP,CTX and NTX were detected by ELISA.Results:H staining examination showed that the bone tissue in the model group showed obvious osteoporotic changes.The pathological changes of osteoporosis in rats were significantly improved after drug intervention;the blood phosphorus and blood calcium concentrations of rats in the model group were lower than those of the blank group(P<0.05),and the blood phosphorus and blood calcium concentrations increased after drug intervention(P>0.05);The concentrations of PINP,PICP,CTX and NTX in rats in the model group were significantly higher than those in the blank group(P<0.05),while the concentrations of PINP,PICP,CTX and NTX in the control group and low,medium and high dose groups were lower than those in the model group.(P<0.05);the concentrations of PINP,PICP,CTX and NTX in rats in the low,medium and high dose groups were lower than those in the control group(P<0.05).There was no statistical difference between groups(P>0.05).Conclusion:Bus
