摘  要:疫苗在流行病预防中发挥了重要作用,重组蛋白疫苗具有廉价足量、安全性良好的优势。作为一种成本效益高、生长迅速、基因操作便捷的宿主,大肠杆菌中已成功实现了重组B群脑膜炎球菌疫苗、重组人乳头瘤病毒疫苗、重组戊型肝炎疫苗等重组蛋白疫苗的生产制备和商业化应用。本文综述了基于大肠杆菌表达系统进行重组蛋白疫苗开发的研究现状,介绍了该宿主中已上市和进入临床阶段的预防性疫苗和治疗性疫苗的研发情况,为大肠杆菌表达系统中重组蛋白疫苗的开发和进一步优化提供理论指导。Vaccines play an important role in epidemic prevention,among which recombinant protein vaccines have the advantages of low cost,sufficient quantity and good safety.As a cost-effective host strain with rapid growth and convenient gene operation,Escherichia coli has successfully achieved the production and commercial application of recombinant protein vaccines such as meningococcal group B vaccine,recombinant human papillomavirus vaccine.This paper reviews the research status of recombinant protein vaccine development based on the E.coli expression system,and introduces the research and development of preventive vaccines and therapeutic vaccines that have been marketed or entered the clinical stage in this host,so as to provide theoretical guidance for the development and further optimization of recombinant protein vaccines in E.coli expression system.
